All hemagglutinin subtypes of IAVs can infect swine; however, only sporadic cases of infection with avian IAVs are reported in domestic swine. The molecular mechanisms affecting the ability of avian IAVs to infect swine are still not fully understood. From the findings of phenotypic analyses, this study suggests that the tissue tropisms (i.e., in swine upper respiratory tracts) of avian IAVs affect their spillovers from wild birds to pigs. It was found that this phenotype is determined not by receptor binding preference but is determined by other markers across genomic segments, such as those in the ribonucleoprotein complex. In addition, our results show that such a phenotypic trait was sporadically and randomly distributed among IAVs across multiple avian species and geographic and temporal orders. Using data on waterfowl band recoveries, we identified spatially explicit hotspots of concentrate... more Using data on waterfowl band recoveries, we identified spatially explicit hotspots of concentrated waterfowl movement to predict occurrence and spatial spread of a novel influenza A virus (clade 2.3.4.4) introduced from Asia by waterfowl from an initial outbreak in North America in November 2014. In response to the outbreak, the hotspots of waterfowl movement were used to help guide sampling for clade 2.3.4.4 viruses in waterfowl as an early warning for the US poultry industry during the outbreak . After surveillance sampling of waterfowl, we tested whether there was greater detection of clade 2.3.4.4 viruses inside hotspots. We found that hotspots defined using kernel density estimates of waterfowl band recoveries worked well in predicting areas with higher prevalence of the viruses in waterfowl. This approach exemplifies the value of ecological knowledge in predicting risk to agricultural security.

Since that time, efforts have been made to improve baiting strategies with a focus on bait density to maximize both efficiency and cost effectiveness. Emerging diseases are increasing burdens on public health, negatively affecting the world economy... more Emerging diseases are increasing burdens on public health, negatively affecting the world economy, causing extinction of species, and disrupting ecological integrity. One Health recognizes that human, domestic animal, and wildlife health are interconnected within ecosystem health and provides a framework for the development of multidisciplinary solutions to global health challenges. To date, most health-promoting interventions have focused largely on single-sector outcomes. For example, risk for transmission of zoonotic pathogens from bush-meat hunting is primarily focused on human hygiene and personal protection. However, bush-meat hunting is a complex issue promoting the need for holistic strategies to reduce transmission of zoonotic disease while addressing food security and wildlife conservation issues. Temporal and spatial separation of humans and wildlife, risk communication, and other preventative strategies should allow wildlife and humans to co-exist. Upstream surveillance, vaccination, and other tools to prevent pathogen spillover are also needed. Clear multi-sector outcomes should be defined, and a systems-based approach is needed to develop interventions that reduce risks and balance the needs of humans, wildlife, and the environment. The ultimate goal is long-term action to reduce forces driving emerging diseases and provide interdisciplinary scientific approaches to management of risks, thereby achieving optimal outcomes for human, animal, and environmental health.

Unlike domestic swine, feral swine are free ranging and have many opportunities for IAV exposure through contacts with various habitats and animals, including migratory waterfowl, a natural reservoir for IAVs. During 2010--2013, 8,239 serum samples were collected from feral swine across 35 US states and tested against 45 contemporary antigenic variants of avian, swine, and human IAVs; of these, 406 (4.9%) samples were IAV-antibody positive. Among 294 serum samples selected for antigenic characterization, 271 cross-reacted with ≥1 testing virus whereas the other 23 did not cross-react with any testing virus. Introductions of low pathogenic avian influenza (LPAI) viruses of subtypes H5 and H7 into poultry... more Introductions of low pathogenic avian influenza (LPAI) viruses of subtypes H5 and H7 into poultry from wild birds have the potential to mutate to highly pathogenic avian influenza (HPAI), but such viruses' origins are often unclear. In January 2016, a novel H7N8 HPAI virus caused an outbreak in turkeys in Indiana, USA. To determine the virus's origin, we sequenced genomes of 441 wild bird-origin influenza A viruses (IAVs) from North America and subjected them to evolutionary analyses. Results showed that the H7N8 LPAI virus most likely circulated among diving ducks in the Mississippi flyway during autumn 2015 and was subsequently introduced to Indiana turkeys, in which it evolved high pathogenicity. Introduced viruses were soon detected in the United States and within six months had spread to 21 states with more than 48 million poultry affected. In an effort to study potential mechanisms of spread of the Eurasian H5 virus, the United States Department of Agriculture coordinated several epidemiologic investigations at poultry farms. As part of those efforts, we sampled synanthropic birds and mammals at five infected and five uninfected poultry farms in northwest Iowa for exposure to avian influenza viruses. Across all farms, we collected 2,627 samples from 648 individual birds and mammals. The One Health concept merges environmental, wildlife, domestic animal, and human health into a g... more The One Health concept merges environmental, wildlife, domestic animal, and human health into a global, interconnected context. Though recently a popular catchphrase due to the emergence and reemergence of diseases such as foot-and-mouth disease (FMD), bluetongue, bovine spongiform encephalopathy (BSE), Q Fever, Rift Valley fever, Nipah virus, West Nile virus (WNV), severe acute respiratory syndrome (SARS), highly pathogenic H5N1 avian influenza, and plague, the One Health concept is not new. The One Health approach to managing infectious diseases aims to promote and implement meaningful collaboration and communication between multiple allied disciplines working locally, nationally, and internationally to attain optimal health for people, domestic animals, wildlife, and our environment. Rabies is an ancient disease that results in more than 55,000 human deaths worldwide each year. Historic and current emphasis on interdisciplinary approaches to rabies control provides a prime example of a zoonotic disease that is being managed more effectively via the One Health approach. In North America, a few meso-carnivores and bats serve as reservoirs of rabies, perpetuating continual infection of humans, pets, and livestock. Increased emphasis on surveillance and control of rabies in wildlife is the key to local, regional, and continental elimination strategies.

In the US, sampling for avian paramyxovirus serotype-1 (APMV-1) is generally conducted when morbi... more In the US, sampling for avian paramyxovirus serotype-1 (APMV-1) is generally conducted when morbidity or mortality events occur involving certain families of wild birds known to be affected by the virus, such as cormorants (Family Phalacrocoracidae), pigeons, doves (Family Columbidae), or pelicans (Family Pelecanidae). To quantify the prevalence of APMV-1 in apparently healthy wild birds and to determine its geographic distribution, we collected swab and serum samples from >3,500 wild birds, representing eight orders from 1 January 2013 to 30 September 2013. Antibody prevalence was highest in wild birds of Order Suliformes (44.9%), followed by Pelecaniformes (24.4%), Anseriformes (22.7%), and Columbiformes (11.7%), with a relatively high occurrence of virulent viruses in Columbiformes (100% of virulent viruses isolated). As expected, viral shedding was comparatively much lower, and positives were only identified in Orders Accipitriformes (1.4%), Columbiformes (1.0%), Anseriformes (0.8%), and Charadriiformes (0.4%). We also demonstrate circulating virulent APMV-1 viruses of genotype VI in apparently healthy Rock Pigeons ( Columba livia ) from March through September in three states.

Wild birds serve as amplifying hosts for many arboviruses, and are thought to be responsible for ... more Wild birds serve as amplifying hosts for many arboviruses, and are thought to be responsible for introducing these viruses into new areas during migration as well as reintroducing them to places where winter temperatures disrupt mosquito-borne transmission. To learn more about four mosquito-borne arboviruses of concern to human or animal health, we tested sera from 997 wild birds of 54 species and 17 families across 44 states of the United States collected from January 1, 2013, through September 30, 2013. Samples were tested for antibody against eastern equine encephalitis, St. Louis encephalitis, West Nile, and Turlock viruses using plaque reduction neutralization tests with an endpoint of 80% or greater. Of the 333 (33.4%) birds that tested positive for antibody to at least one arbovirus, 29.7% were exposed to two or more arboviruses. Exposure to all four arboviruses was detected in Canada geese, double-crested cormorants, mallards, mute swans, laughing gulls, and American coots. Our results suggest that exposure to arboviruses is widespread in the United States across a diversity of wild bird species.

Introductions of low pathogenic avian influenza (LPAI) viruses of subtypes H5 and H7 into poultry... more Introductions of low pathogenic avian influenza (LPAI) viruses of subtypes H5 and H7 into poultry from wild birds have the potential to mutate to highly pathogenic avian influenza (HPAI), but such viruses' origins are often unclear. In January 2016, a novel H7N8 HPAI virus caused an outbreak in turkeys in Indiana, USA. To determine the virus's origin, we sequenced genomes of 441 wild bird-origin influenza A viruses (IAVs) from North America and subjected them to evolutionary analyses. Results showed that the H7N8 LPAI virus most likely circulated among diving ducks in the Mississippi flyway during autumn 2015 and was subsequently introduced to Indiana turkeys, in which it evolved high pathogenicity. Introduced viruses were soon detected in the United States and within six months had spread to 21 states with more than 48 million poultry affected. In an effort to study potential mechanisms of spread of the Eurasian H5 virus, the United States Department of Agriculture coordinated several epidemiologic investigations at poultry farms. As part of those efforts, we sampled synanthropic birds and mammals at five infected and five uninfected poultry farms in northwest Iowa for exposure to avian influenza viruses. Though recently a popular catchphrase due to the emergence and reemergence of diseases such as foot-and-mouth disease (FMD), bluetongue, bovine spongiform encephalopathy (BSE), Q Fever, Rift Valley fever, Nipah virus, West Nile virus (WNV), severe acute respiratory syndrome (SARS), highly pathogenic H5N1 avian influenza, and plague, the One Health concept is not new. The One Health approach to managing infectious diseases aims to promote and implement meaningful collaboration and communication between multiple allied disciplines working locally, nationally, and internationally to attain optimal health for people, domestic animals, wildlife, and our environment. Rabies is an ancient disease that results in more than 55,000 human deaths worldwide each year. Historic and current emphasis on interdisciplinary approaches to rabies control provides a prime example of a zoonotic disease that is being managed more effectively via the One Health approach. In North America, a few meso-carnivores and bats serve as reservoirs of rabies, perpetuating continual infection of humans, pets, and livestock. Increased emphasis on surveillance and control of rabies in wildlife is the key to local, regional, and continental elimination strategies.

In the US, sampling for avian paramyxovirus serotype-1 (APMV-1) is generally conducted when morbi... more In the US, sampling for avian paramyxovirus serotype-1 (APMV-1) is generally conducted when morbidity or mortality events occur involving certain families of wild birds known to be affected by the virus, such as cormorants (Family Phalacrocoracidae), pigeons, doves (Family Columbidae), or pelicans (Family Pelecanidae). To quantify the prevalence of APMV-1 in apparently healthy wild birds and to determine its geographic distribution, we collected swab and serum samples from >3,500 wild birds, representing eight orders from 1 January 2013 to 30 September 2013. Antibody prevalence was highest in wild birds of Order Suliformes (44.9%), followed by Pelecaniformes (24.4%), Anseriformes (22.7%), and Columbiformes (11.7%), with a relatively high occurrence of virulent viruses in Columbiformes (100% of virulent viruses isolated). As expected, viral shedding was comparatively much lower, and positives were only identified in Orders Accipitriformes (1.4%), Columbiformes (1.0%), Anseriformes (0.8%), and Charadriiformes (0.4%). We also demonstrate circulating virulent APMV-1 viruses of genotype VI in apparently healthy Rock Pigeons ( Columba livia ) from March through September in three states.

Wild birds serve as amplifying hosts for many arboviruses, and are thought to be responsible for ... more Wild birds serve as amplifying hosts for many arboviruses, and are thought to be responsible for introducing these viruses into new areas during migration as well as reintroducing them to places where winter temperatures disrupt mosquito-borne transmission. To learn more about four mosquito-borne arboviruses of concern to human or animal health, we tested sera from 997 wild birds of 54 species and 17 families across 44 states of the United States collected from January 1, 2013, through September 30, 2013. Samples were tested for antibody against eastern equine encephalitis, St. Louis encephalitis, West Nile, and Turlock viruses using plaque reduction neutralization tests with an endpoint of 80% or greater. Of the 333 (33.4%) birds that tested positive for antibody to at least one arbovirus, 29.7% were exposed to two or more arboviruses. Exposure to all four arboviruses was detected in Canada geese, double-crested cormorants, mallards, mute swans, laughing gulls, and American coots. Our results suggest that exposure to arboviruses is widespread in the United States across a diversity of wild bird species.
