In apple trees (Malus domestica), cold temperatures induce bud dormancy at the end of summer/beginning of the fall. Apple buds stay dormant during winter until they are exposed to a period of cold known as a chilling requirement. Once the chilling requirement is satisfied, dormancy is released, and buds can resume growth (budbreak) and initiate flowering in response to warm temperatures in spring. In the annual model plant species Arabidopsis thaliana, the thermo-regulation of flowering-time is strongly controlled by the epigenetic and post-transcriptional modification of floral-regulators genes mediated by small RNAs. Small RNAs modulate temperature responses also in woody plant species, such as grapevine, poplar, and pear. However, how small RNAs are involved in genetic networks of temperature-mediated to regulate dormancy and budbreak in fruit tree species remains unclear. To study the role of small RNAs on the control of apple dormancy, we performed a small RNA sequencing on bud samples during a time-course experiment. Using the method “TraPR” (Trans-kingdom, rapid, affordable Purification of RISCs), 373 different potential micro RNAs (miRNAs) were identified, including 105 known apple miRNAs. A total of 39 of them were differentially expressed during the dormancy cycle, suggesting their role in this process. The possible targets were identified in silico and classified according to their potential molecular function. Remarkably, targets of the identified miRNAs were related to growth control and hormonal signaling, cell wall modification, global transcriptional and post-transcriptional regulation, among other significant pathways. In conclusion, we identified several apple miRNAs and their targets potentially involved in genetic networks controlling dormancy and budbreak in fruit tree species

In Arabidopsis thaliana, the photoperiodic pathway acts through FLOWERING LOCUS T (FT) to promote floral induction in response to day length. A complex between FT and the bZIP transcription factor FD is proposed to form in the SAM leading to activation of APETALA1 (AP1) and LEAFY (LFY) and thereby promoting floral meristem identity. We identified mutations that suppress FT function and recovered a new allele of the homeodomain transcription factor PENNYWISE (PNY). Genetic and molecular analyses showed that ectopic expression of BLADE-ON-PETIOLE1 (BOP1) and BOP2, which encode transcriptional co-activators, in the SAM during vegetative development confers the late flowering of pny mutants. In wild-type plants BOP1/2 are expressed in lateral organs close to boundaries with the SAM, whereas in pny mutants their expression occurs in the SAM. This ectopic expression lowers FD mR...

Seasonal cues of day length or winter cold trigger flowering of many species. Forward and reverse... more Seasonal cues of day length or winter cold trigger flowering of many species. Forward and reverse genetic approaches are revealing the mechanisms by which these responses are conferred. Homologues of the Arabidopsis thaliana protein FLOWERING LOCUS T (FT) are widely used to mediate seasonal responses to day length and act as graft-transmissible promoters or repressors of flowering. Winter cold in A. thaliana promotes flowering by repressing transcription of the MADS box gene FLOWERING LOCUS C (FLC). The mechanism by which this occurs involves a complex interplay of different forms of long noncoding RNAs induced at the FLC locus during cold and changes in the chromatin of FLC. In perennial relatives of A. thaliana, flowering also requires the age-dependent downregulation of miRNA156 before winter.

The pervaporation performance was tested on the of waterethanol mixtures in the range 85-96 wt. % ethanol. The permeate flux increased from 0.45 to 0.55 kg m-2 h-1 at 50 ºC for the ETS-10/CS MMM with respect to the pure CS membranes. Characterization by SEM and TEM, XRD, DSC and TGA allowed inferring an intimate contact between the dispersed ETS-10 and the continuous chitosan phase. The 5 wt. % loading of titanosilicate scarcely decreased the hydrophilic character of the mixed matrix membrane but increased the molecular sieving effect on the transport and separation properties, thus affecting the membrane behavior on pervaporation.

Flowering is the best characterized of these seasonal responses, and in temperate climates it often occurs in spring. Genetic approaches in Arabidopsis thaliana have shown how the underlying responses to changes in day length (photoperiod) or winter temperature (vernalization) are conferred and how these converge to create a robust seasonal response. Recent advances in plant genome analysis have demonstrated the diversity in these regulatory systems in many plant species, including several crops and perennials, such as poplar trees. Here, we report progress in defining the diverse genetic mechanisms that enable plants to recognize winter, spring and autumn to initiate flower development.

The main objective of this study is to determine the frequency of MDD in persons aged 65 years and older using the Detection of Depression in the Elderly Scale (DDES). A second objective is to determine the convergent validity of the DDES with the Geriatric Depression Scale (GDS).Methods: A cross-sectional, observational study was carried out of 1,387 subjects aged 65 years and older. The variables considered were: affective state (GDS and DDES), physical and cognitive functional state, health problems and sociodemographic characteristics. Results: Using the DDES we identified MDD in 50 subjects (4.3%). There was a moderate correlation (r = 0.570; p < 0.001) between the DDES and the GDS scores (p < 0.001). According to logistic regression analysis, the variables associated with a probable MDD (DDES +) were: dependence in activities of daily living ...

An observational study was carried out for the evaluation of the Insomnia in the Elderly Scale using the psychiatric interview as the gold standard. Data was collected in a primary care setting in the city of Albacete, Spain. Participants were 926 non-institutionalized older adults aged ≥ 65 years. Sleep characteristics, and the health and sociodemographic status of the participants were assessed. The Insomnia in the Elderly Scale is divided into two subscales for the evaluation of the Diagnostic and Statistical Manual of Mental Disorders, Fouth Edition criteria A and B for insomnia. The area under the receiver operating characteristic curve for subscale A was 0.868. The cut-off point to detect the presence of diagnostic criterion A for insomnia was a score of ≥ 3 (sensitivity: 86.4%; specificity: 69.5%). The area under the receiver operating characteristic curve for subscale B was 0.832. The cut-off point to detect the presence of diagnostic criterion B was a score of ≥ 2 (sensitivity: 86.3%; specificity: 66.4%). The Insomnia in the Elderly Scale has appropriate psychometric properties. This scale compensates for the lack of validated instruments for use in the population aged 65 years or older. The fact that it is divided into two subscales to separately evaluate the insomnia criteria enables us to detect the presence of each of them. Finally, it has been validated in participants aged 65 years and older, which is the target population for this scale.
