Sipahutar pineapple from North Sumatra, Indonesia, is considered difficult to breed, resulting in... more Sipahutar pineapple from North Sumatra, Indonesia, is considered difficult to breed, resulting in a lack of seed availability. However, tissue culture allows the emergence of somaclonal variations, especially in plantlets undergoing several subculture stages. This study aimed to identify somaclonal variations in Sipahutar pineapple seedlings generated from in vitro culture using Inter Simple Sequence Repeat (ISSR) markers. Fifteen accessions were used in this study consisting of Sipahutar pineapple grown in its original environment, a laboratory environment, acclimatized, and in vitro cultured. Total of 10 primers, of which seven primers detected polymorphisms among the population. This study resulted in a low genetic similarity among the Sipahutar pineapple population. The genetic diversity of Sipahutar pineapple plants propagated by in vitro culture had a similarity level of 75%-94%, showing somaclonal variations in vitro culture. The genetic similarity of pineapple accessions was divided into two main clusters, namely cluster A consisted of 11 accessions and cluster B consisted of 4 accessions. Cluster separation does not correlate to the source of plant materials studied.

It aimed to isolate and identify sponge symbiont bacteria with antibacterial potential against Multi-Drug Resistance Organisms (MDRO) with the 16S rRNA gene marker. Based on the study results, two sponges were obtained, namely Clathrina sp and Agelas sp, each having 11 and 7 isolates of symbiont bacteria, respectively. Based on the antibacterial activity test results, out of 18 isolates of symbiont bacteria, only 4 had antibacterial potential against MDRO (Klebsiella pneumoniae ESBL, Pseudomonas aeruginosa ESBL, or Staphylococcus lugdunensis MRSA) with inhibition zones of 9.3 mm, 8.3 mm, and 8.1 mm, respectively. The results of bacterial identification using the 16S rRNA gene sequence isolate S1I3, S1I9, and A1I3 belong to the Bacillus cereus and Bacillus paramycoides types. This research is expected to provide important information about sponge symbiont bacteria with antibacterial potential against MDRO.

This study aims to analyze students' digital literacy in the context of learning biology mate... more This study aims to analyze students' digital literacy in the context of learning biology material on the digestive system and identify students' digital literacy in terms of interest in digital technology, internet costs, gender, and learning outcomes. The research was conducted at one private high school in Medan, Indonesia. The instrument used is a questionnaire and a list of student grades from the teacher. The results showed that students interested in digital technology had a significantly higher digital literacy score than students who had less interest in digital technology. There was no significant difference in digital literacy between groups of students with internet fees < US$ 14 per month and students with internet fees of US$ 14 per month, and between groups of male and female students. The result of the analysis also found a positive correlation between learning outcomes and digital literacy.

Lichen is a symbiotic organism consisting of algae (photobionts) and fungi (mycobiont). In Indone... more Lichen is a symbiotic organism consisting of algae (photobionts) and fungi (mycobiont). In Indonesia, Usnea is the only genus of lichen that has been widely recognized and used by the local community for traditional medicine. It is locally known as kayu angin, rusuk angin, or janggot Kai. Furthermore, it produces lichen acid and other compounds as secondary metabolites that need investigation. Therefore, this study aimed to identify species and determine the secondary metabolites of Usnea in West Java. There were 13 species of Usnea identified, with U. rubrotincta, U. mutabilis, and U. barbata being new to West Java. The extraction of U. esperantiana has the highest number of lichen acids, with 20 different types identified. This study showed that Usnea contains lichen acid, flavonoid, and saponin compounds. The secondary metabolites are a potential source of natural products for bioprospecting.

Control of Xoo bacteria can be accomplished through the use of Xoo-resistant rice varieties. Due to the shifting genetic structure and environmental conditions of Xoo bacteria, it is difficult to identify them according to their pathotype. This study aims to determine the genetic diversity of Xoo bacteria using the polymerase chain reaction (PCR) method and the IS1112-based RAPD marker, which is known to contain repetitive sequences. DNA was isolated from 52 isolates collected from 15 districts and cities in North Sumatra province and then used to amplify the IS1112 sequence. Dendrogram analysis revealed that cluster analysis of PCR findings classified isolates into 15 groups with a 90% similarity value. In the territory of Indonesia, andaliman is spread in the highlands of North Sumatra and parts of Aceh. Andaliman is a spice plant that is usually used as a spice for the Batak tribe in North Sumatra. This study aims to analyze the characteristics of andaliman habitat in North Sumatra. A total of 169 samples from 8 regencies in North Sumatra analyzed habitat characteristics using the ArcGis 10.3 program using various ecological data. The results showed that andaliman was widely distributed in the highlands in North Sumatra. Zanthoxylum acanthopodium is a member of Rutaceae family widely found in northern Sumatra, Indonesia. The aim of this study was to barcode Z. acanthopodium in North Sumatra province, Indonesia based on cpDNA maturase K (matK). Samples were collected in seven localities across six regions of North Sumatra province. Phylogenetic analysis was conducted using Maximum Likelihood method. The results of phylogenetic analysis indicate that Z. acanthopodium is a monophyletic group that is derived from a common ancestor. The results of the phylogenetic tree construction show that there is a grouping of accession between Z. acanthopodium species separate from other species in the Zanthoxylum genus as well as those of the Rutaceae family. The results showed that cpDNA matK marker can effectively be used as DNA barcoding to identify Z. acanthopodium.

This species is commonly called the Lesser Whistling-duck because it flies in a noisy sound. In Indonesia, it is known as Belibis Batu. Dendrocygna javanica is spread in the regions of Asia including Indonesia. In Indonesia, D. javanica is spread on the islands of Sumatra, Kalimantan, Java, East Nusa Tenggara and West Nusa Tenggara. This study aims to create a distribution model of Dendrocygna javanica based on habitat characteristics in North Sumatra, Indonesia, using GIS (Geographic Information System). A total of 35 distribution points of D. javanica in 8 districts in North Sumatra were analyzed using ArcGis 10.3 software to analyze habitat characteristics. The results showed that D. javanica spread on habitat characteristics Rainfall ranged from 1500-3500 mm/year, 5 soil types namely ferric acrisols (4 points), humic acrisols (4 points), dystric cambisols (2 points), orthic ferrasols (5 point), and dystric fluvisols (20 points), 7 types of land cover namely dryland forest (2 points), plantation (5 points), dryland agriculture (10 points), rice fields (2 points), swamp (4 points), plus (4 points), and body of water (8 points), and are at an elevation of 3-1072 meters above sea level. Endophytic microbial colonization is found in various types of plant organs, namely: roots, stems, leaves and seeds. This study is focused on endophytic bacteria mostly conducted intensively on wild plants or forests in the tropical ecosystems. Various trees or tropical forest plants have potentials as traditional medicine and in cooperation with the bacteria. The C. melanoxylon has been utilized by local community as antidiarrheal medicine because of the content of active antibacterial compounds. The presence of endophytic bacteria in tissues of the stem of C. melanoxylon was observed the bacteria were directly isolated from the tissues by planting pour plate on Tryptic Soya Agar (TSA) medium. The antibacterial activity of endophytic bacteria isolates was examined using a dual cultural assay against Escherichia coli ATCC 35218 and Staphyllococcus aureus ATCC 25923 as a model bacterial. Morphological, physiological, and biochemical characters were identified based on the ability of the isolates to grow. The genetic similarity of pineapple accessions was divided into two main clusters, namely cluster A consisted of 11 accessions and cluster B consisted of 4 accessions. It aimed to isolate and identify sponge symbiont bacteria with antibacterial potential against Multi-Drug Resistance Organisms (MDRO) with the 16S rRNA gene marker. Based on the study results, two sponges were obtained, namely Clathrina sp and Agelas sp, each having 11 and 7 isolates of symbiont bacteria, respectively. Based on the antibacterial activity test results, out of 18 isolates of symbiont bacteria, only 4 had antibacterial potential against MDRO (Klebsiella pneumoniae ESBL, Pseudomonas aeruginosa ESBL, or Staphylococcus lugdunensis MRSA) with inhibition zones of 9.3 mm, 8.3 mm, and 8.1 mm, respectively. The results of bacterial identification using the 16S rRNA gene sequence isolate S1I3, S1I9, and A1I3 belong to the Bacillus cereus and Bacillus paramycoides types. This research is expected to provide important information about sponge symbiont bacteria with antibacterial potential against MDRO.

This study aims to analyze students' digital literacy in the context of learning biology mate... more This study aims to analyze students' digital literacy in the context of learning biology material on the digestive system and identify students' digital literacy in terms of interest in digital technology, internet costs, gender, and learning outcomes. The research was conducted at one private high school in Medan, Indonesia. The instrument used is a questionnaire and a list of student grades from the teacher. The results showed that students interested in digital technology had a significantly higher digital literacy score than students who had less interest in digital technology. There was no significant difference in digital literacy between groups of students with internet fees < US$ 14 per month and students with internet fees of US$ 14 per month, and between groups of male and female students. The result of the analysis also found a positive correlation between learning outcomes and digital literacy.

In Indonesia, Usnea is the only genus of lichen that has been widely recognized and used by the local community for traditional medicine. It is locally known as kayu angin, rusuk angin, or janggot Kai. Furthermore, it produces lichen acid and other compounds as secondary metabolites that need investigation. Therefore, this study aimed to identify species and determine the secondary metabolites of Usnea in West Java. There were 13 species of Usnea identified, with U. rubrotincta, U. mutabilis, and U. barbata being new to West Java. The extraction of U. esperantiana has the highest number of lichen acids, with 20 different types identified. This study showed that Usnea contains lichen acid, flavonoid, and saponin compounds. The secondary metabolites are a potential source of natural products for bioprospecting.

Zanthoxylum acanthopodium is a member of Rutaceae family widely found in northern Sumatra, Indonesia. The aim of this study was to barcode Z. acanthopodium in North Sumatra province, Indonesia based on cpDNA maturase K (matK). Samples were collected in seven localities across six regions of North Sumatra province. Phylogenetic analysis was conducted using Maximum Likelihood method. The results of phylogenetic analysis indicate that Z. acanthopodium is a monophyletic group that is derived from a common ancestor. The results of the phylogenetic tree construction show that there is a grouping of accession between Z. acanthopodium species separate from other species in the Zanthoxylum genus as well as those of the Rutaceae family. The results showed that cpDNA matK marker can effectively be used as DNA barcoding to identify Z. acanthopodium.

This species is commonly called the Lesser Whistling-duck because it flies in a noisy sound. In Indonesia, it is known as Belibis Batu. Dendrocygna javanica is spread in the regions of Asia including Indonesia. In Indonesia, D. javanica is spread on the islands of Sumatra, Kalimantan, Java, East Nusa Tenggara and West Nusa Tenggara. This study aims to create a distribution model of Dendrocygna javanica based on habitat characteristics in North Sumatra, Indonesia, using GIS (Geographic Information System). A total of 35 distribution points of D. javanica in 8 districts in North Sumatra were analyzed using ArcGis 10.3 software to analyze habitat characteristics. The results showed that D. javanica spread on habitat characteristics Rainfall ranged from 1500-3500 mm/year, 5 soil types namely ferric acrisols (4 points), humic acrisols (4 points), dystric cambisols (2 points), orthic ferrasols (5 point), and dystric fluvisols (20 points), 7 types of land cover namely dryland forest (2 points), plantation (5 points), dryland agriculture (10 points), rice fields (2 points), swamp (4 points), plus (4 points), and body of water (8 points), and are at an elevation of 3-1072 meters above sea level. Endophytic microbial colonization is found in various types of plant organs, namely: roots, stems, leaves and seeds. This study is focused on endophytic bacteria mostly conducted intensively on wild plants or forests in the tropical ecosystems. Various trees or tropical forest plants have potentials as traditional medicine and in cooperation with the bacteria. The C. melanoxylon has been utilized by local community as antidiarrheal medicine because of the content of active antibacterial compounds. The presence of endophytic bacteria in tissues of the stem of C. melanoxylon was observed the bacteria were directly isolated from the tissues by planting pour plate on Tryptic Soya Agar (TSA) medium. The antibacterial activity of endophytic bacteria isolates was examined using a dual cultural assay against Escherichia coli ATCC 35218 and Staphyllococcus aureus ATCC 25923 as a model bacterial. Tanaman ini memiliki variasi fenotip karena keragaman genetik dan adaptasi di berbagai ragam kondisi lingkungan. Tujuan dari penelitian ini adalah mengklasifikasikan Bouea macrophylla Griffit berdasarkan variasi genetik menggunakan DNA kloroplas. Daerah TrnL-F intergenic spacer merupakan daerah nonkoding pada DNA kloroplas yang dapat digunakan sebagai penanda untuk melihat variasi genetik intraspesifik. Sampel diperoleh dari 5 tempat di Indonesia yaitu Ambon, Banten, Aceh, Sumatera Barat, dan Kalimantan Barat. Isolasi DNA menggunakan metode Doyle dan Doyle yang dimodifikasi, kemudian diamplifikasi menggunakan primer trnL-F forward dan primer trnL-F reverse. Produk PCR dengan panjang fragmen 350-400 bp kemudian disekuensing di 1st Base Singapura. Data hasil sekuensing dianalisis menggunakan software MEGA 6.06 untuk membangun pohon filogenetik menggunakan out group Mangifera indica dan Anacardium occidentale. Hasil dari peneltian ini menunjukkan bahwa daerah trnL-F pada DNA kloroplas adalah daerah dengan konservatif yang tinggi. Bouea macrophylla Griffit memiliki kedekatan dengan Anacardium occidentael. Analisis pohon filogenetik dengan menggunakan Software Mega 6.06 menunjukkan bahwa konsistensi spesies tersebut dari nenek moyang yang sama dan memisah dari spesies Mangifera indica dan Anacaardium occidentale sebagai out group.
