Juveniles of Prochilodus lineatus were exposed to sediments collected from one of five sites alon... more Juveniles of Prochilodus lineatus were exposed to sediments collected from one of five sites along an urban stream into which various types of contaminants are discharged. After 24 or 96 h fish were examined and the results compared with those from control groups (fish exposed only to water, for the same period). Plasma ion levels varied significantly and fish exposed to site 5 sediment showed a transient increase in both sodium and chloride concentrations. Plasma glucose was significantly higher in fish exposed to sediment from sites 2 and 5. The higher liver glutathione-S-transferase activity registered in fish exposed to sediment from sites 1, 4 and 5 suggests the presence of organic contaminants at these sites and the enhancement of liver catalase activity in fish exposed to sediment from sites 3 and 4 may be due to contaminant-mediated oxyradical production. The overall results revealed that sites 4 and 5 are more severely contaminated, probably due to organic contaminants from agricultural sources and municipal landfill.

The objective of this work was to evaluate the effects of this herbicide on the Neotropical fish Prochilodus lineatus. Juvenile fish were acutely exposed (6, 24 and 96 h) to 1 mg L À1 of RDT (RDT 1), 5 mg L À1 of RDT (RDT 5) or only water (control) and blood samples for hematological analysis, liver for antioxidants analysis, and brain and muscle for acetylcholinesterase (AChE) determination, were collected. RDT effects were more evident in fish exposed to the higher concentration of the herbicide. Hematologic alterations appeared only after 96 h exposure, when fish showed an increase in the hematocrit and in the number of both red and white blood cells. After 6 h exposure fish showed a transient reduction in superoxide dismutase and catalase activity. RDT also inhibited glutathione-S-transferase, after 6 and 24 h of exposure. The reduction in these enzymes is probably related to the occurrence of lipid peroxidation (LPO) in fish exposed to the herbicide for 6 h. LPO returned to control levels after 24 and 96 h exposure to RDT, when fish showed an increased activity of glutathione peroxidase. The content of reduced glutathione also increased after 96 h exposure. Thus, after 24 and 96 h the antioxidant defenses were apparently enough to combat ROS, preventing the occurrence of oxidative damage. The exposure to RDT for 96 h led to an inhibition of AChE in brain and muscle at rates which may not be considered a life-threatening situation.

Fish were acutely exposed (6, 24 and 96 h) to 10 mg L À1 of Roundup Ò (RD) or only water (control) and samples of liver, for antioxidants analysis, and brain and muscle, for acetylcholinesterase (AChE) determination, were collected. Fish exposed to RD for 24 h showed reduction on superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities, and increased glutathione (GSH) content. After 24 and 96 h, fish of RD group showed increased glutathione-S-transferase (GST) activity and lipid peroxidation. AChE activity was inhibited in brain after 96 h and in muscle after 24 and 96 h of exposure. Thus, acute exposure to RD stimulated the biotransformation pathway, with increased GST, but interfered on the antioxidant defenses, with reduction of SOD and GPx activity, leading to the occurrence of lipid peroxidation. Inhibition of AChE showed that RD acts as a contaminant with anti-AChE action.

This study evaluated the osmo- and ionic balance, Na/K-ATPase, H-ATPase and carbonic anhydrase activities and the mitochondria-rich cells (MRC) in the gills and kidney of the Neotropical fish Prochilodus lineatus after 2 (acute) and 14 (subchronic) days of exposure to nominal 0, 1, 5, 10 and 50 mg L TiO-NP. This phenotype promotes a high degree of protection against xenobiotics. The present study provides a better understanding of the MXR activity in the podal disk cells of Bunodosoma cangicum exposed to copper, and further establishes the relationship between protein activity (measured by accumulation of rhodamine-B) and bioaccumulation of copper in these cells. Sea anemone cells were exposed for 24h to copper (0, 7.8 and 15.6μg/L) in presence and absence of MXR blocker (verapamil 50μM). Results indicate that copper exposure increases intracellular metal content when ABC proteins were blocked, causing an increase in cellular death. Preliminary series of short-term static toxicity tests were run to determine LC50 (96 h) of lead in P. lineatus, which was 95 mg Pb.L-1. Therefore, lead concentrations tested in the sublethal experiments were 24 and 71 mg Pb.L-1, which correspond to 25% and 75% of the LC50 (96 h), respectively. Gills of P. lineatus exposed to both lead concentrations during 96 h presented a higher occurrence of histopathological lesions such as epithelial lifting, hyperplasia, and lamellar aneurism. P. lineatus did not show significant alterations in hematocrit during exposure to both lead concentrations. Since atrazine can have adverse effects on non-target crops, here we analyzed the effect of encapsulated atrazine on growth, physiological and oxidative stress parameters of soil-grown maize plants (Zea mays L.). Poly(epsilon-caprolactone) (PCL) nanocapsules have been recently developed as a modified release ... more Poly(epsilon-caprolactone) (PCL) nanocapsules have been recently developed as a modified release system for atrazine, an herbicide that can have harmful effects in the environment. Here, the post-emergence herbicidal activity of PCL nanocapsules containing atrazine was evaluated using mustard (Brassica juncea) as target plant species model. Characterization of atrazine-loaded PCL nanocapsules by nanoparticle tracking analysis indicated a concentration of 7.5 x 10 12 particles mL-1 and an average size distribution of 240.7 nm. The treatment of mustard plants with nanocapsules carrying atrazine at 1 mg mL-1 resulted in a decrease of net photosynthesis and PSII maximum quantum yield, and an increase of leaf lipid peroxidation, leading to shoot growth inhibition and the development of severe symptoms. Time course analysis until 72 h after treatments showed that nanoencapsulation of atrazine enhanced the herbicidal activity in comparison with a commercial atrazine formulation. In contrast to the commercial formulation, tenfold dilution of the atrazine-containing nanocapsules did not compromise the herbicidal activity. No effects were observed when plants were treated with nanocapsules without herbicide compared to control leaves sprayed with water. Overall, these results demonstrated that atrazine-containing PCL nanocapsules provide very effective post-emergence herbicidal activity. More importantly, the use of nanoencapsulated atrazine enables the application of lower dosages of the herbicide, without any loss of efficiency, which could provide environmental benefits.

In the present study, an integrated approach examining genotoxic, biochemical and physiological parameters was employed to evaluate acute effects of POEA on the Neotropical fish Prochilodus lineatus. Juvenile fish were exposed to 0.15 mg·L(-1) (POEA 1), 0.75 mg·L(-1) (POEA 2) and 1.5 mg·L(-1) (POEA 3) of POEA or only water (CTR), and after 24h exposure samples of blood and liver were taken. In fish caged in the urban stream the most common lesions were epithelial lifting, hyperplasia and hypertrophy of the respiratory epithelium, lamellar fusion, and aneurysms in the gills; enlargement of the glomerulus, reduction of Bowman's space, occlusion of the tubular lumen, cloudy swelling and hyaline droplet degeneration in the kidneys; hepatocytes with hypertrophy, cytoplasmic and nuclear degeneration, melanomacrophage aggregates, bile stagnation and one case of focal necrosis in the liver. Applying an integrated approach using the Comet, micronucleus (MN), and random amplified polymorp... more Applying an integrated approach using the Comet, micronucleus (MN), and random amplified polymorphic DNA (RAPD) assays, occurrence of erythrocytic nuclear abnormalities (ENAs) and the liver activity of antioxidants enzymes (catalase and glutathione-S-transferase (GST)) was carried out to evaluate the effects of acute (6, 24, and 96 h) and subchronic (15 days) exposures to aluminum on fish Prochilodus lineatus. The Comet assay showed that fish erythrocytes exhibited significantly higher DNA damage after 6 and 96 h of Al exposure. MN frequencies were very low and did not increase significantly after Al exposures, while ENAs frequency increased significantly after all exposure periods. RAPD profiles obtained with DNA from fish fins collected before the toxicity tests were compared to the profiles with DNA from gills and liver of the same fish sampled after Al exposures. Alterations in RAPD profiles, including appearance and disappearance of bands, after 6 h, 24 h, and 15 days of Al exposure were detected. Fish exposed to Al for 6 and 24 h also showed significant increases in GST and catalase activities. These results indicated that Al exposure was genotoxic to P. lineatus, inducing DNA damage in peripheral erythrocytes. The induction of antioxidant enzymes might be an indication that Al causes oxidative damage to DNA, while the very low frequency of MN suggests that Al does not produce clastogenic or aneugenic effects. Genotoxic effects after 15 days of Al exposure was revealed only by RAPD, showing that this assay represents a sensitive method to detect genotoxic damage, occasionally not detected by other genotoxic tests used in toxicological genetics studies.

In the present study, random amplified polymorphic DNA (RAPD) markers were used to analyze genetic variability and structure of populations assigned to the Neotropical fish species Astyanax scabripinnis from an urban stream located in Londrina, Parana´State, southern Brazil. Thirty individuals of this species were collected from three sites throughout the upper Cambe´stream. A total of 10 primers amplified 159 loci, of which 128 (80.5%) were polymorphic. Each of the three populations showed very similar proportions of polymorphic loci, which ranged from 63.5 to 64.8%. Unbiased genetic distances varied from 0.0612 to 0.0646. Theta p-test values indicated moderate to high genetic differentiation among individuals from different localities. The number of migrants varied from 1.34 to 1.46, suggesting a low gene flow between populations. The genetic similarity among all individuals studied ranged from 0.424 to 0.848. The results suggest that populations of A. scabripinnis in Cambe´stream are undergoing genetic differentiation.

In this study, random amplified polymorphic DNA (RAPD) markers were applied to analyze the geneti... more In this study, random amplified polymorphic DNA (RAPD) markers were applied to analyze the genetic diversity of samples of the Neotropical catfish Hypostomus ancistroides, collected from four sites (S1, S2, S3 and S4) along an urban stream in Southern Brazil. The 11 primers used in RAPD analysis amplified 147 loci, 76 (51.7%) of which were polymorphic. The proportions of polymorphic loci observed in the four samples were: 29.93% (S1), 31.97% (S2), 23.81% (S3) and 38.77% (S4). The average heterozygosity within sampling localities ranged from 0.1230 to 0.1526 and unbiased genetic distances ranged from 0.0253 to 0.0445. AMOVA partitioned 90.85% of the total variation within samples and 9.15% among samples. Excepting for the sample pair S1-S2 (φ ST = 0.02784; p > 0.05), all others pairwise φ ST values were significantly greater than zero, indicating moderate genetic differentiation among catfish samples from four localities. The relative low levels of genetic variation detected in all groups studied could be related to different factors, including the sedentary habit of these fish, which can be eroding the genetic variation of H. ancistroides from each locality.

Juvenile fish were exposed to Pb (from 1.7 to 0.7 mg of dissolved Pb L-1) for 6, 24 and 96 h and Pb accumulation was analyzed in the gills, liver, kidneys, blood cells and muscle. The following parameters were also analyzed: hematologic (hemoglobin content, hematocrit and number of red blood cells), metabolic (blood glucose), endocrine (blood cortisol), osmo ionic (plasma osmolality and Na ? , K ? , Cland Ca ?2 concentrations), gill enzymes (Na ? /K ?-ATPase and carbonic anhydrase), chloride cell (CC) density and CC location in the gills. Pb accumulated in all the analyzed tissues, with the kidneys showing the highest concentration, followed by the gills and liver. The lowest Pb concentrations were found in blood cells and muscle. Pb promoted an increase in blood glucose after 6 and 24 h exposure. Gill Na ? /K ?-ATPase was inhibited after 24 h of exposure, but its activity was restored at 96 h, probably due to the increase in CC in gill lamellae. Plasma Na ? was reduced after 6 and 96 h, while K ? concentrations increased at all the experimental times. Fish exposed to Pb showed reduced plasma Ca ?2 at all experimental periods. Hematologic parameters remained unchanged. Overall, this study demonstrated that Pb interferes in osmoregulatory processes of P. lineatus and the proliferation of CC in the gills is a response in order to reestablish adequate ion concentrations. After 24 or 96 h fish were examined and the results compared with those from control groups (fish exposed only to water, for the same period). Plasma ion levels varied significantly and fish exposed to site 5 sediment showed a transient increase in both sodium and chloride concentrations. Plasma glucose was significantly higher in fish exposed to sediment from sites 2 and 5. The higher liver glutathione-S-transferase activity registered in fish exposed to sediment from sites 1, 4 and 5 suggests the presence of organic contaminants at these sites and the enhancement of liver catalase activity in fish exposed to sediment from sites 3 and 4 may be due to contaminant-mediated oxyradical production. The overall results revealed that sites 4 and 5 are more severely contaminated, probably due to organic contaminants from agricultural sources and municipal landfill.

The objective of this work was to evaluate the effects of this herbicide on the Neotropical fish Prochilodus lineatus. Juvenile fish were acutely exposed (6, 24 and 96 h) to 1 mg L À1 of RDT (RDT 1), 5 mg L À1 of RDT (RDT 5) or only water (control) and blood samples for hematological analysis, liver for antioxidants analysis, and brain and muscle for acetylcholinesterase (AChE) determination, were collected. RDT effects were more evident in fish exposed to the higher concentration of the herbicide. Hematologic alterations appeared only after 96 h exposure, when fish showed an increase in the hematocrit and in the number of both red and white blood cells. After 6 h exposure fish showed a transient reduction in superoxide dismutase and catalase activity. RDT also inhibited glutathione-S-transferase, after 6 and 24 h of exposure. The reduction in these enzymes is probably related to the occurrence of lipid peroxidation (LPO) in fish exposed to the herbicide for 6 h. LPO returned to control levels after 24 and 96 h exposure to RDT, when fish showed an increased activity of glutathione peroxidase. The content of reduced glutathione also increased after 96 h exposure. Thus, after 24 and 96 h the antioxidant defenses were apparently enough to combat ROS, preventing the occurrence of oxidative damage. The exposure to RDT for 96 h led to an inhibition of AChE in brain and muscle at rates which may not be considered a life-threatening situation.

Fish were acutely exposed (6, 24 and 96 h) to 10 mg L À1 of Roundup Ò (RD) or only water (control) and samples of liver, for antioxidants analysis, and brain and muscle, for acetylcholinesterase (AChE) determination, were collected. Fish exposed to RD for 24 h showed reduction on superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities, and increased glutathione (GSH) content. After 24 and 96 h, fish of RD group showed increased glutathione-S-transferase (GST) activity and lipid peroxidation. AChE activity was inhibited in brain after 96 h and in muscle after 24 and 96 h of exposure. Thus, acute exposure to RD stimulated the biotransformation pathway, with increased GST, but interfered on the antioxidant defenses, with reduction of SOD and GPx activity, leading to the occurrence of lipid peroxidation. Inhibition of AChE showed that RD acts as a contaminant with anti-AChE action.

Preliminary series of short-term static toxicity tests were run to determine LC50 (96 h) of lead in P. lineatus, which was 95 mg Pb.L-1. Here, the post-emergence herbicidal activity of PCL nanocapsules containing atrazine was evaluated using mustard (Brassica juncea) as target plant species model. Characterization of atrazine-loaded PCL nanocapsules by nanoparticle tracking analysis indicated a concentration of 7.5 x 10 12 particles mL-1 and an average size distribution of 240.7 nm. The treatment of mustard plants with nanocapsules carrying atrazine at 1 mg mL-1 resulted in a decrease of net photosynthesis and PSII maximum quantum yield, and an increase of leaf lipid peroxidation, leading to shoot growth inhibition and the development of severe symptoms. Time course analysis until 72 h after treatments showed that nanoencapsulation of atrazine enhanced the herbicidal activity in comparison with a commercial atrazine formulation. In contrast to the commercial formulation, tenfold dilution of the atrazine-containing nanocapsules did not compromise the herbicidal activity. No effects were observed when plants were treated with nanocapsules without herbicide compared to control leaves sprayed with water. Overall, these results demonstrated that atrazine-containing PCL nanocapsules provide very effective post-emergence herbicidal activity. More importantly, the use of nanoencapsulated atrazine enables the application of lower dosages of the herbicide, without any loss of efficiency, which could provide environmental benefits.

Applying an integrated approach using the Comet, micronucleus (MN), and random amplified polymorp... more Applying an integrated approach using the Comet, micronucleus (MN), and random amplified polymorphic DNA (RAPD) assays, occurrence of erythrocytic nuclear abnormalities (ENAs) and the liver activity of antioxidants enzymes (catalase and glutathione-S-transferase (GST)) was carried out to evaluate the effects of acute (6, 24, and 96 h) and subchronic (15 days) exposures to aluminum on fish Prochilodus lineatus. The Comet assay showed that fish erythrocytes exhibited significantly higher DNA damage after 6 and 96 h of Al exposure. MN frequencies were very low and did not increase significantly after Al exposures, while ENAs frequency increased significantly after all exposure periods. RAPD profiles obtained with DNA from fish fins collected before the toxicity tests were compared to the profiles with DNA from gills and liver of the same fish sampled after Al exposures. Alterations in RAPD profiles, including appearance and disappearance of bands, after 6 h, 24 h, and 15 days of Al exposure were detected. Fish exposed to Al for 6 and 24 h also showed significant increases in GST and catalase activities. These results indicated that Al exposure was genotoxic to P. lineatus, inducing DNA damage in peripheral erythrocytes. The induction of antioxidant enzymes might be an indication that Al causes oxidative damage to DNA, while the very low frequency of MN suggests that Al does not produce clastogenic or aneugenic effects. Genotoxic effects after 15 days of Al exposure was revealed only by RAPD, showing that this assay represents a sensitive method to detect genotoxic damage, occasionally not detected by other genotoxic tests used in toxicological genetics studies.

In the present study, random amplified polymorphic DNA (RAPD) markers were used to analyze genetic variability and structure of populations assigned to the Neotropical fish species Astyanax scabripinnis from an urban stream located in Londrina, Parana´State, southern Brazil. Thirty individuals of this species were collected from three sites throughout the upper Cambe´stream. A total of 10 primers amplified 159 loci, of which 128 (80.5%) were polymorphic. Each of the three populations showed very similar proportions of polymorphic loci, which ranged from 63.5 to 64.8%. Unbiased genetic distances varied from 0.0612 to 0.0646. Theta p-test values indicated moderate to high genetic differentiation among individuals from different localities. The number of migrants varied from 1.34 to 1.46, suggesting a low gene flow between populations. The genetic similarity among all individuals studied ranged from 0.424 to 0.848. The results suggest that populations of A. scabripinnis in Cambe´stream are undergoing genetic differentiation.

In this study, random amplified polymorphic DNA (RAPD) markers were applied to analyze the geneti... more In this study, random amplified polymorphic DNA (RAPD) markers were applied to analyze the genetic diversity of samples of the Neotropical catfish Hypostomus ancistroides, collected from four sites (S1, S2, S3 and S4) along an urban stream in Southern Brazil. The 11 primers used in RAPD analysis amplified 147 loci, 76 (51.7%) of which were polymorphic. The proportions of polymorphic loci observed in the four samples were: 29.93% (S1), 31.97% (S2), 23.81% (S3) and 38.77% (S4). The average heterozygosity within sampling localities ranged from 0.1230 to 0.1526 and unbiased genetic distances ranged from 0.0253 to 0.0445. AMOVA partitioned 90.85% of the total variation within samples and 9.15% among samples. Excepting for the sample pair S1-S2 (φ ST = 0.02784; p > 0.05), all others pairwise φ ST values were significantly greater than zero, indicating moderate genetic differentiation among catfish samples from four localities. The relative low levels of genetic variation detected in all groups studied could be related to different factors, including the sedentary habit of these fish, which can be eroding the genetic variation of H. ancistroides from each locality.

Juvenile fish were exposed to Pb (from 1.7 to 0.7 mg of dissolved Pb L-1) for 6, 24 and 96 h and Pb accumulation was analyzed in the gills, liver, kidneys, blood cells and muscle. The following parameters were also analyzed: hematologic (hemoglobin content, hematocrit and number of red blood cells), metabolic (blood glucose), endocrine (blood cortisol), osmo ionic (plasma osmolality and Na ? , K ? , Cland Ca ?2 concentrations), gill enzymes (Na ? /K ?-ATPase and carbonic anhydrase), chloride cell (CC) density and CC location in the gills. Pb accumulated in all the analyzed tissues, with the kidneys showing the highest concentration, followed by the gills and liver. The lowest Pb concentrations were found in blood cells and muscle. Pb promoted an increase in blood glucose after 6 and 24 h exposure. Gill Na ? /K ?-ATPase was inhibited after 24 h of exposure, but its activity was restored at 96 h, probably due to the increase in CC in gill lamellae. Plasma Na ? was reduced after 6 and 96 h, while K ? concentrations increased at all the experimental times. Fish exposed to Pb showed reduced plasma Ca ?2 at all experimental periods. Hematologic parameters remained unchanged. Overall, this study demonstrated that Pb interferes in osmoregulatory processes of P. lineatus and the proliferation of CC in the gills is a response in order to reestablish adequate ion concentrations.
