Study on outbreak of Neospora caninum-associated abortion in dairy cows in Tabriz (Northwest Iran) by serological, molecular and histopathologic methods

To determine Neospora caninum (N. caninum) as a cause of bovine abortion in dairy cows by ELISA, PCR and Pathological methods in Tabriz, Northwest of Iran. For study of outbreak of neosporosis, blood samples were collected from 76 Holstein aborted dairy cows in Tabriz (Northwest Iran). Antibodies to N. caninum were assayed by using a commercially ELISA kit (IDEXX, USA). IgG against N. caninum were found in 14 (18.4%) cases. Aborted fetuses of these seropositive dams were proposed for histopathological and molecular investigations. Brains, spinal cords and placentas of the fetuses were fixed in 10% buffered formalin for histopathology. Also 5-10 g of brain tissue was sampled for DNA extraction. In 6 out of 14 (42.8%) fetuses, brain tissue was positive in PCR. All dams of these fetuses were serologically seropositive in ELISA test. SEROLOGICAL SCREENING FOR Neospora caninum INFECTION IN A DAIRY CATTLE FARM WITH REPRODUCTIVE DISORDERS, IN SOUTH-EASTERN ROMANIA

Neospora caninum is an apicomplexan protozoa parasite that causes reproductive disorders in cattle worldwide. Despite the high seroprevalence reported in Romania, there are limited data on N. caninum infection in dairy cattle, and no studies have evaluated the presence of infection over time in a limited area. Therefore, this study aimed to assess the seroprevalence of N. caninum in a dairy cattle farm in Southeastern Romania where a previously study reported about the cattle exposure to N. caninum infection. A total of 85 cattle, assigned in four groups, according to the reproductive status (repeated breeding syndrome, history of abortion, pregnant cows, and fresh cows) were tested for the presence of N. caninum antibodies, using an indirect ELISA test. The overall seroprevalence was 28.23% (95% CI: 18.67-37.80). According to the reproductive groups, the seroprevalence varied between 19.04% (95% CI: 2.25-35.84) in fresh cows to 38.09% (95% CI: 17.32-58.87) in cows with repeated breeding syndrome. By these findings new information is provided about the disease's presence in the studied farm, implying that infection with N. caninum may be the primary cause of reported reproductive disorders, as well as about the infection's persistent nature.

Introduction Neospora caninum is an apicomplexan protozoon causing abortion in cattle worldwide. Infected cows represent economic losses due to fetal abortion, stillbirth, clinical and subclinical disease, impaired milk production, neonatal deaths, reduced fertility and reduced value of infected cows. Neospora caninum Suspects as One of the Most Important Causes of Abortion in Large Dairy Farms in Isfahan, Iran

Background: This study aimed to reveal the serological prevalence of Neospora caninum in large dairy farms in Isfahan Province, central Iran. Methods: Serum samples were collected from 1500 cattle living in four large dairy farms in Isfahan Province, Iran during 2014–2015 and examined for anti N. caninum IgG antibodies. Overall, 113 serum samples were also collected from the dogs living in these areas; suspecting to be risk factors for this infection. All the serum samples were investigated to find IgG antibodies by using ELISA. Dogs’ sera were also analyzed by indirect fluorescent antibody test. Results: Totally, 395 out of 1500 bovine samples (26.33%) were positive for N. caninum: 34%, 21.61%, 23.03% and 29.01% in four investigated clusters (farms). Prevalence of Neospora caninum infection in dairy cows and its consequences for reproductive management

This study was carried out to determine the prevalence of neosporosis in an area of intensive dairy production, in Portugal. Sera samples were obtained in a random basis from 114 cows in 49 herds (group A), and from 1237 cows in 36 herds with a history of abortion outbreaks (group B). All sera samples were tested for neosporosis by direct agglutination test (DAT). Additionally, attempts to isolate Neospora caninum in 42 aborted bovine fetuses from 38 dairy herds (group C) were carried out, utilizing a bioassay with immuno-depressed Swiss Webster mice. Parasitological confirmation was done by indirect fluorescent antibody test (IFAT). The prevalence of neosporosis in the group A was 28%. Group B had a significantly (P < 0:001) higher prevalence (46%) and Neospora caninum was isolated in 36% of the aborted fetuses (group C). These results indicate that neosporosis, a disease only recently (2001) diagnosed in Portugal, has a high prevalence in the country, particularly in populations with a story of abortion. Thus, neosporosis should systematically be considered in the differential diagnosis of abortion. In the context of embryo transfers, the importance of selecting Neospora-free embryo recipients is discussed, as well as the pertinence of assessing the Neospora status of traded and imported cattle.

Seroprevalence of Neospora caninum in dairy cows in Belgrade city area, Serbia

The protozoan parasite Neospora caninum is one of the most important abortifacient pathogen in cattle. Serological investigations are often used in order to estimate seroprevalence in herds. Aims of our study were to determine the seroprevalence among aborting and non-aborting dairy cattle in Belgrade city area as well as epidemiological factors that are important for N. caninum infection. Using commercial ELISA kit, we examined 188 sera. Out of 188, 142 samples originated from pregnant (non-aborting) cows from five farms, while 46 were from cows who had aborted. Overall seroprevalence was 25% (48/188). Seroprevalence was significantly higher (p≤0.05) in aborting than in non-aborting group of cows (37% and 21.1% respectively). Relation between Neospora caninum and abortion in dairy cows: Risk factors and pathogenesis of disease

Neosporosis is a parasitic disease cause by Neospora caninum, a parasite of great importance in livestock. This study aimed to evaluate the presence of antibody against N. caninum in dairy cattle with history of abortion, as well as to identify associated risk factors for neosporosis. Animals suspected of neosporosis (n = 130) after clinical examination were randomly selected. Sera samples from 29 farms were submitted to indirect immunofluorescence technique (IFA) in order to detect antibodies against N. caninum, and animals were considered positive if ≥IFA 1:200. An epidemiological questionnaire was used to verify probable risk factors for neosporosis and their cause-effect relation. Seroepidemiology of Neospora caninum infection in dairy cattle herds in Mashhad area, Iran

A seroepidemiology study of Neospora caninum was conducted in Mashhad area, Khorasan province, Iran on 337 cattle from 30 dairy using ELISA. All herds had seropositive cattle and antibodies were formed in 46% of the cattle. Abortion was significantly associated with seropositivity of cattle. The first study on seroprevalence and risk factors of Neospora caninum infection in pregnant local cows from Northeast Algeria

Background and Aim: Neospora caninum is one of the most common infectious organisms worldwide that causes abortion in cattle. To the best of our knowledge, no previous studies have focused on N. caninum infection in the local Atlas brown cattle from Northeast Algeria. This study aimed to assess the importance of bovine neosporosis for causing abortion in Atlas brown cattle and to identify selected risk factors. Materials and Methods: A case-control study was performed on 60 control farms and 30 case farms. We collected 650 blood samples from 650 pregnant cows from 90 farms in five Algerian provinces; Jijel, Skikda, Annaba, El-Tarf, and Souk- Ahras. Sera samples were analyzed for the presence of antibodies against N. caninum using enzyme-linked immunosorbent assays. Results: The seroprevalence of N. caninum infection in the cows was 36.2% (95% confidence interval [CI]: 32.7-39.8) and in the farms was 81.1% (95% CI: 73.0-89.2). Risk factors found by multivariable logistic regression included: Presence of dogs (odds ratio [OR] 4.7, 95 CI 2.9-7.3); age ≥84 months (OR 4.9, 95 CI 2.8-8.3); Jijel region (OR 2.2, 95 CI 1.1-4.5); white (OR 2.5, 95 CI 1.4-4.4) and gray (OR 2.5, 95 CI 1.4-4.5) coat; moderate (OR 2.30, 95 CI 1.4-3.8) and bad (OR 3.1, 95 CI 1.8-5.3) hygiene; and second (OR 2.5, 95 CI 1.4-4.4); and last (OR 2.3, 95 CI 1.3-4.2) stage of pregnancy. Our case-control study showed no significant association between seropositivity of N. caninum and abortion at the farms level (OR 0.9, 95 CI 0.3-2.7). Similarly, there was no significant association between seropositivity of N. caninum and abortion at the individual level (OR 0.8, 95 CI 0.6-1.2). Conclusion: This is the first study of N. caninum infection in pregnant local cows from Northeast Algeria. The prevalence rate of antibodies against N. caninum was high. Almost all risk factors studied for infection were significantly associated with seroprevalence. Our analysis showed no relation between N. caninum infection and abortion. Consequently, these local cows are resistant to abortion caused by N. caninum.

The Seroprevalence of Neospora Caninum Antibodies in Dairy Cattle Herds in Golestan Province, Iran

A total of 800 sera collected from six herds of health and aborted cows in Golestan province in Iran (579 without abortion history, 221 with abortion history) were examined for presence of Neospora caninum antibodies by using commercial ELISA kit. The overall seroprevalence of Neospora caninum antibodies was 107 from 800 (13.37±2.36%, α=0.05). Comparison of Neospora caninum serological status in age groups (<2 years, 2-4 years, >4 years) showed no significant difference (p>0.05). The prevalence of Neospora caninum was higher in the aborted cows than in non aborted cows. This is the first study on Neospora caninum antibodies in dairy cattle herds in Golestan province in the north east of Iran.
