Evaluation of the antioxidant activity as an additional parameter to attain the functional quality of natural extracts

Due to differences in the functional quality of natural extracts, we have also faced differences in their effectiveness. So, it was intended to assess the antioxidant activity of natural extracts in order to attain their functional quality. It was observed that all the extracts (brown and green propolis, Ginkgo biloba and Isoflavin Beta®) and the standard used (quercetin) showed antioxidant activity in a dose-dependent manner with IC 50 values ranging from 0.21 to 155.28 μg mL-1 (inhibition of lipid peroxidation and scavenging of the DPPH • assays). We observed a high correlation (r 2 = 0.9913) among the antioxidant methods; on the other hand, the antioxidant activity was not related to the polyphenol and flavonoid content. As the DPPH • assay is a fast method, presents low costs and even has a high correlation with other antioxidant methods, it could be applied as an additional parameter in the quality control of natural extracts. RESUMEN. "Evaluación de la actividad antioxidante como parámetro adicional para lograr la calidad funcional de extractos naturales". Debido a las diferencias en la calidad funcional de extractos naturales, nosotros hemos advertido también diferencias en su efectividad, por lo que se pretende estimar la actividad antioxidante de extractos naturales para lograr su calidad funcional. Fue observado que todos los extractos (propolis marrón y verde, Ginkgo biloba e Isoflavin Beta®) y el patrón usado (quercetina) mostraron actividad antioxidante de una manera dosis-dependiente con valores de IC 50 que van de 0,21 a 155,28 al μg mL-1 (inhibición de la peroxidación lipídioca y captación de radicales libres del DPPH •). Se observó una alta correlación (r 2 = 0,9913) entre los métodos antioxidantes y por otro lado la actividad antioxidante no estuvo relacionada con el contenido del polifenoles ni de flavonoides. Como el análisis del DPPH • es un método rápido, presenta costos bajos e incluso tiene una correlación alta con otros métodos antioxidantes, este método podría utilizarse como un parámetro adicional en el control de calidad de extractos naturales.

Assessment of antioxidant capacity of some extracts for further use in therapy

Salviae and Plantago species are plants known as having antioxidant effects. The aim of the study was to compare in vitro the antioxidant capacity of these medicinal plants from two different areas of Romania (Bihor and Arad counties) in order to use it in the future in the field of pharmacology and medicine. An UV-Vis spectra fingerprint (250-800 nm) of ethanol extracts from Salviae and Plantago species was recorded in order to establish the maximum absorption, specifically bioactive compounds. The bioactive compounds from those two extracts, polyphenols types were determined by the Folin-Ciocalteu method, and the total flavonoids have been determined through a spectrophotometric method. Antioxidant capacity of the extracts was evaluated by the following methods: DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2'-Azinobis [3-ethylbenzothiazoline-6-sulfonic acid]-diammonium salt) and FRAP (the Ferric reducing antioxidant power). The results shown, that Salviae species are rich in polyphenols, while Plantago is rich in flavonoids being a good reason for their association.

The anti-oxidative activity of plant-derived extracts is well-known and confers health-promoting effects on functional foods and food supplements. Aim of this work is to evaluate the capability of two different assays to predict the real biological antioxidant efficiency. At this purpose, extracts from five different plant-derived matrices and commercial purified phytochemicals were analyzed for their anti-oxidative properties by using well-standardized in vitro chemical method (TEAC) and an ex vivo biological assay. The biological assay, a cellular membrane system obtained from erythrocytes of healthy volunteers, is based on the capability of phytochemicals treatment to prevent membrane lipid peroxidation under oxidative stress by UV-B radiation. Antioxidants in biological system have multiple functions; which includes, protection from oxidative damage caused by free radicals, generated during the redox reactions in the physiological environment. There are several preliminary screening methodologies available for evaluation of antioxidant activity which includes DPPH radical scavenging, ABTS scavenging, hydroxyl radical scavenging, hydrogen peroxide scavenging, super oxide scavenging, nitric oxide scavenging etc. These methods are mainly in vitro models for evaluation of antioxidant potential of test samples and are found to be efficient in predicting antioxidant activity. In this review, we revisited and reviewed the different in vitro methods available for antioxidant activity measurement.

Effect of Extraction Solvent/Technique on the Antioxidant Activity of Selected Medicinal Plant Extracts

The effects of four extracting solvents [absolute ethanol, absolute methanol, aqueous ethanol (ethanol: water, 80:20 v/v) and aqueous methanol (methanol: water, 80:20 v/v)] and two extraction techniques (shaking and reflux) on the antioxidant activity of extracts of barks of Azadirachta indica, Acacia nilotica, Eugenia jambolana, Terminalia arjuna, leaves and roots of Moringa oleifera, fruit of Ficus religiosa, and leaves of Aloe barbadensis were investigated. The tested plant materials contained appreciable amounts of total phenolic contents (0.31-16.5 g GAE /100g DW), total flavonoid (2.63-8.66 g CE/100g DW); reducing power at 10 mg/mL extract concentration (1.36-2.91), DPPH . scavenging capacity (37.2-86.6%), and percent inhibition of linoleic acid (66.0-90.6%). Generally higher extract yields, phenolic contents and plant material antioxidant activity were obtained using aqueous organic solvents, as compared to the respective absolute organic solvents. Although higher extract yields were obtained by the refluxing extraction technique, in general higher amounts of total phenolic contents and better antioxidant activity were found in the extracts prepared using a shaker.

Antioxidants are the substances that are capable of counteracting the damaging effects of the physiological process of oxidation occur in animal tissues. These may be nutrients (vitamins and minerals) as well as enzymes (proteins) in our body that assist in chemical reactions. They are believed to play a role in preventing the development of chronic diseases like cancer, diabetes, heart disease, stroke, Alzheimer's disease, rheumatoid arthritis, and cataracts etc. The present review article emphasizes on the various aspects of oxidants and antioxidants viz. definition, types, causes, mechanism, functions, adverse effects along with various in vivo and in vitro models of evaluation of antioxidant activity of new molecule, compounds or any plant or plant part. This review is one of its kinds which will be of great importance for researchers working in this area in search of antioxidant moieties and their biological evaluation.

In-vitro antioxidant and GC-MS analysis ethanolic extract of poly herbal drug

Antioxidants play an important role in inhibiting and scavenging free radicals, thus providing protection to human against infections and degenerative diseases. Current research is now directed towards natural antioxidants originated from plants due to safe therapeutics. Poly herbal drugs is used in Indian traditional medicine for a wide range of various ailments. To understand the mechanism of pharmacological actions, antioxidant properties of the Poly herbal drugs extract were tested using standard in vitro models. The ethanolic extract of Poly herbal drugs exhibited strong scavenging effect on superoxide, nitric oxide radical and reducing power radical scavenging assay. The free radical scavenging effect of Poly herbal drugs extract was comparable with that of the reference antioxidants. The data obtained in the present study suggests that the extract of Poly herbal drugs have potent Invitro antioxidant and Anti Diabetic activity against free radicals, prevent oxidative damage to major biomolecules and afford significant protection against oxidative damage.
