This study aimed to determine the effect of extract of tomato (Solanum lycopersicum L.) on the in... more This study aimed to determine the effect of extract of tomato (Solanum lycopersicum L.) on the incidence of mammary gland ductal histology changes in a carcinogenic substance-7,12-dimetilbenz(α)antrasena (DMBA- induced mice and the optimal dose of tomato extract to reduce the effect of DMBA. For that, this study used 30 female mice which were grouped into 6 treatment groups (K+, K-, P1, P2, P3, P4). K+ was control group which did not receive treatment, K- was control group with DMBA without the extract of tomato, while the P1, P2, P3, and P4 treated with DMBA and extracts tomatoes, respectively, with each dose was 100, 250, 400 and 500 mg/kgBW given fourth a week for six week. The dose of DMBA was 0,56 mg/20gBW that was given in corn oil twice a week for 5 weeks. All treatments administered orally.
At the end of treatment, mammary glands were collected, fixed, processed into histological section and stained with hematoxylin eosin. The obtained data were diameter, lumen diameter, epithelium thickness and epithelia cell layer types of the mammary gland ducts. They were analyzed by one-way ANOVA followed by Duncan test but the data of lumen diameter duct by Brown Forsythe test followed by Games-Howell test (α = 0,05). The lycopene level of tomato was measured by spectrophotometer method, whereas antioxidant activity of tomato was done by DPPH scavenging method. The results showed that the extract of tomato at doses of 400 and 500 mg/kgBW could reduce lumen diameter and increase ductal epithelium thickness. Optimum dose of tomato extract was 500 mg/kgBW.

Samples were collected from Madura strait, East Java, Indonesia, each month for February 2012 until January 2013. Reproductive peak was predicted using data of gonadal index. Gonadal index is ratio of gonad to body wall weight. Gonadal cycle was determined by observed maturity stages of gonad. Maturity stages, sex determination, and identification of gametogenic cell types were identified histologically on 4 μm thick cross section of gonadal tubules. Spermatozoa morphology was observed by scanning electron microscope. The results showed that there was no reproductive peak of P. australis, but the reproductive activity increased during dry season. P. australis is dioecious with sex ratio approaching 1:1. Gonadal maturation stages were observed as gametogenesis, mature, spawning, and post-spawning. Gametogenic cells in gonadal tubules were identified as spermatogonium, spermatocyte, spermatid, and spermatozoa in male, and previtelogenic, vitelogenic, and mature oocyte in female

Is the first research conducted in Indonesia. This study aims to measure the n-hexane extract of the body wall of sea cucumber P. dobsoni which is related to the maturity level of the gonads. The measurement results obtained that the average wet weight and dry weight of sea cucumber body wall was 23.58 grams of body wall wet weight, body wall dry weight 5.56, gonad weight 4.02, and GSI value 18.03. Judging from the histology of male sea cucumber gonads, it is known that they have grown and grown at an advanced stage with an average content of n-hexane extract at an average value of 0.202 grams and 0.266 grams, respectively. Whereas in the female group there are all stages of growth, continued growth, maturity, recovery and post-spawning. It was done experimentally on male mice (strain BALB, aged 8–10 weeks, weighed 25–35 g). Thirty eight mice were grouped to 4 (four) groups: First group was treated with solvent (as positive control), 2nd group was treated ethynilestradiol (EE2) (as negative control), 3rdgroup was treated with Java ginseng root extract, and 4th group was treated with Korean ginseng root extract. All groups were administered with EE2 0.56 mg/20 g bw/day for 9 days as pretreatment to decrease the testosteron level. After pretreatment, each group divided to 3 subgroups (each would receive treatment for 9, 18, and 27 days). An alloxan injection was used to induce diabetic condition (0.13 mg/g mice weight) via intraperitoneal injection. After the topical photosensitizer injection was done, the wounds were exposed with laser beam on the first day the incision wound has been made. Each wound length was measured every day with millimeter ruler. For the wound width, it was measured after Hematoxylin and Eosin staining. Red Okra (Abelmoschus esculentus Moench) is widely distributed in Indonesia and its antioxidant a... more Red Okra (Abelmoschus esculentus Moench) is widely distributed in Indonesia and its antioxidant ability is unknown. This study was performed to evaluate the antioxidant activities of red okra pods extracts. The powder of red okra was extracted by reflux. The first extraction used n-hexane (three times). Ethyl acetate was used to extract the residue (three times). Then, the residue was extracted three times using ethanol. Antioxidant activities were tested using DPPH and FRAP assays. This study showed that ethanol extract showed the highest antioxidant activity, which had the lowest IC50 DPPH (39.8 µg/mL) and the highest FRAP (89.15 µmol Fe2+/g). The result of DPPH and FRAP methods indicates that ethanol extract of red okra pods is classified as a very strong antioxidant.

This study uses male rats (Rattus novergicus), aged 8-12 weeks weighing 150-200 grams, with a total of 31 rats. Therapy was conducted by putting rats in a chamber that emits an electric field every day for 1 hour for 28 days. To search for the optimum frequency of the treatment, frequency variations of 15, 30, 60, and 120 kHz were given. The electrical charge distribution of the air in the room was then measured, whereas mice blood sugar levels were also measured to reveal the effect of the treatment on cell tolerance to sugar, and to come up with the measurement of insulin levels at the end of the treatment. The flavonoid and total phenolic levels, serum biochemistry, and liver histology were examined. Materials and Methods: Green okra pod extraction was performed using ethanol methanol solvent. Thirty adult male BALB/c mice (8-10 weeks, ∼30 g) were divided into six groups: Normal control, negative control (sodium nitrite 50 mg/kg BW exposure), and treatment groups (sodium nitrite exposure and OPME at doses of 50, 100, 200, and 400 mg/kg BW). This study aims to evaluate the effects of alginate extract from Sargassum ilicifolium and okra (... more This study aims to evaluate the effects of alginate extract from Sargassum ilicifolium and okra (Abelmoschus esculentus) pods extract on the re-epithelialization rate of the wound area and the number of neutrophils, macrophages, fibroblasts, fibrocytes, and collagen density in streptozotocin-induced diabetic mice. This study was done in vivo using male mice (strain BALB/C, 3 to 4 months old, 30 to 40g). The normal control group (KN) and the diabetic groups (induced by streptozotocin). The diabetic group was divided into 9 groups: three diabetic control groups (KD), three alginate treatment groups (KAI), and three treatment groups of alginate-okra (Abelmoschus esculentus) pods extract (KAIO). The treatment dose was 50 mg/kg body weight. Measurement of fasting blood sugar levels was done before and after streptozotocin injection. The treatment was carried out in three different time periods: 3 days, 7 days, and 14 days. Then, mice skin was cut and proceed for histopathological analysis. Interestingly, we indicated that the administration of okra (Abelmoschus esculentus) pods extract combined with alginate from Sargassum ilicifolium could increase the rate of re-epithelialization of the wound area, increase the number of neutrophils, macrophages, fibroblasts, fibrocytes, as well as increase synthesized collagen. The combined extract was able to improve the healing process of open wounds in diabetic mice. Therefore, it can be concluded that the administration of alginate from Sargassum ilicifolium and okra (Abelmoschus esculentus) pods extract was able to improve open wounds healing in diabetic mice due to the role of antioxidant from polysaccharides and quercetin which acted as anti-hyperglycemic to anti-inflammatory properties.

However, only few studies have been reported immunomodulatory activities of okra to prevent disease caused by bacteria infection. In this study, the immunomodulatory activities of polysaccharides from okra pods were investigated further through the spleen weight and B-lymphocytes proliferation in mice infected by Staphylococcus aureus. Okra polysaccharides were obtained by water extraction and ethanol precipitation. Okra polysaccharides with doses of 25, 50, 75, and 100 mg/kg BW were orally administrated to mice with or without Staphylococcus aureus infection. Spleen weight was evaluated in both treatment group and control group. B-lymphocytes proliferation was evaluated by MTT assay using LPS induction. The purpose of this study was to determine the effect of various fractions of okra pods extract (... more The purpose of this study was to determine the effect of various fractions of okra pods extract (VOPE) on superoxide dismutase (SOD) level and tissue glucose tolerance in the streptozotocin (STZ) induced diabetic mice. This study was using 35 male Balb/c strain mice. The samples were divided into seven groups, namely KN (normal control), KD (diabetic control), KA (acarbose control), EK (crude extract), NP (non-polar), SP (semi-polar), and EP (polar). VOPE and acarbose suspension were administered orally for 14 days in the STZ-induced diabetic mice. Intraperitoneal injection of multiple low-dose STZ (30mg/kg body weight) induced diabetes daily for five consecutive days. On the 15th day, the mice were terminated. Data of SOD level and tissue glucose tolerance were analyzed by ANOVA, at a = 0.05. This study concluded that the administration of VOPE could increase SOD level and tissue glucose tolerance in the STZ-induced diabetic mice. In conclusion, VOPE is a promising antioxidant agent due to its antihyperglycemic properties.

In this study, we used male mice (Mus musculus) of the BALB/C strain which were divided into 2 groups: the control group (without treatment of mangosteen pericarp extract) and the treatment group. The control group was divided into three: normal control (KN), diabetic control (KD), diabetic control-Metformin HCl (KM). The treatment group (with mangosteen pericarp extract) was divided into 3 groups (P1, P2, and P3) with the dose of 50 mg/kg body weight, 100 mg/kg body weight, and 200 mg/kg body weight, respectively. The induction of diabetes was done with the injection of multiple low-doses of STZ (30 mg/kg of body weight) for 5 consecutive days. Before and after STZ injection, random blood glucose and total cholesterol were measured at 1 st , 7 th , and 14 th day of mangosteen pericarp extract treatments. Treatments were given for 14 days. At 15 th day, SGOT, SGPT and plasma creatinine levels were measured using Pentra C200, while liver and kidney were collected and then processed into histological slides. Interestingly, we found that mangosteen pericarp extract administration was able to reduce random blood glucose, total cholesterol, SGOT, SGPT, plasma creatinine levels, also ameliorates damaged liver hepatocytes and renal proximal tubular cells in diabetic mice significantly. In conclusion, mangosteen pericarp extract is a promising antidiabetic agent due to its anti-hyperglycemic and antioxidant properties.

In this study, we used male BALB/C mice. Mice were divided into two groups: normal control (KN) and streptozotocin-induced diabetic. Streptozotocin (STZ) induction was performed using multiple low doses of 30 mg/kg body weight injected for five consecutive days. The diabetic mice were separated into three subgroups: diabetic control (KD), diabetic mice treated with acarbose (KA), and diabetic mice treated with gamma-mangostin at either 0,5 mg/kg body weight (P1), 1 mg/kg body weight (P2), or 2 mg/kg body weight (P3). Before and after STZ injection, fasting blood glucose and cholesterol level would be observed. Fasting blood glucose and cholesterol level were also measured at the 1 st , 7 th , and 14 th day of gamma-mangostin treatment. Treatment was given for 14 days. On the 15 th day, SGOT and SGPT were measured using a Pentra C 200 Reader, while the liver was collected and processed onto the histological slides. Interestingly, we found that gamma-mangostin was able to reduce the fasting blood glucose, cholesterol, SGOT, SGPT, and ameliorate the damaged hepatocytes in significant diabetic mice. Therefore, we concluded that gamma-mangostin is a promising anti-diabetic agent due to its anti-hyperglycemic and antioxidant activities.

This study employed 36 male Balb/c strain mice. The samples were divided into seven groups, KN (normal control), KD (diabetic control), KA (acarbose control), EK (crude extract), NP (non-polar), SP (semi-polar), and EP (polar). VOPE and acarbose were administered orally for 14 days in streptozotocin-induced type-2 diabetic mice. Mice with type-2 diabetes mellitus were induced by the intraperitoneal injection of multiple low-dose streptozotocin (30 mg/kg body weight) daily for five consecutive days. On 15th day, mice were terminated. The data of fasting blood glucose, serum insulin, and GLUT4 density were analyzed and interestingly it showed that the administration of VOPE could ameliorate the fasting blood glucose, serum insulin, and GLUT4 density. Therefore, it can be suggested that VOPE can be regarded as a promising antidiabetic agent in the streptozotocin-induced diabetic mice.

The study aimed to determine the hepatoprotective effect of crude polysaccharides extracted from Ganoderma lucidum against liver injury induced by carbon tetrachloride (CCl4). Materials and Methods: Twenty-four male BALB/C mice were randomly divided into six groups. Serum and liver samples were taken on day 10 after G. lucidum administration. The levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), malondialdehyde (MDA), superoxide dismutase (SOD), and catalase (CAT) were measured using enzyme-linked immunosorbent assays, and the histology of the liver was evaluated using light microscopy. Results: G. lucidum extract significantly decreased the levels of ALT, AST, and MDA and significantly increased the levels of SOD and CAT. It can activate granulocyte, monocyte, and macrophage. Administration of crude polysaccharide of C. versicolor against bacterial infection is hypothesized to increase immune response. This study assessed the crude polysaccharides activity of C. versicolor in improving immune response after induction of non-tuberculosis mycobacteria, Mycobacterium fortuitum. Animal model was female mice strain BALB/c aged 8-10 weeks. A crude polysaccharide of C. versicolor was administered before and/or after the bacteria infection for 10 days at a dose of 50 mg/kg body weight. Mice exposure to M. fortuitum was performed twice at a dose of 0.5 Mc. Farland. After crude polysaccharides treatment, both serum and peritoneal fluid were isolated. All data collected were analyzed statistically using ANOVA and Duncan test. Oral administration of crude polysaccharide was found to increase phagocyte number (P<0.05, from crude polysaccharides administration before-after infection), improve phagocytic activity (P<0.05, from crude polysaccharides administration before infection), raise both IFN- and antibody level (P<0.05, from crude polysaccharides administration after and before-after infection), and it caused TNF-α levels to tend to a normal concentration (P>0.05, TNF-α levels after crude polysaccharides administration were relatively the same as controls). Administration of crude polysaccharides of C. versicolor could enhance non-specific immune response, specific immune response, and pro-inflammatory cytokine in mice infected by M. fortuitum. These results suggested that crude polysaccharides of C. versicolor may be applied as an effective immunostimulatory agent.

An alloxan injection was used to induce diabetic condition (0.13 mg/g mice weight) via intraperitoneal injection. After the topical photosensitizer injection was done, the wounds were exposed with laser beam on the first day the incision wound has been made. Red Okra (Abelmoschus esculentus Moench) is widely distributed in Indonesia and its antioxidant a... more Red Okra (Abelmoschus esculentus Moench) is widely distributed in Indonesia and its antioxidant ability is unknown. This study was performed to evaluate the antioxidant activities of red okra pods extracts. The powder of red okra was extracted by reflux. The first extraction used n-hexane (three times). Ethyl acetate was used to extract the residue (three times). Then, the residue was extracted three times using ethanol. Antioxidant activities were tested using DPPH and FRAP assays. This study showed that ethanol extract showed the highest antioxidant activity, which had the lowest IC50 DPPH (39.8 µg/mL) and the highest FRAP (89.15 µmol Fe2+/g). The result of DPPH and FRAP methods indicates that ethanol extract of red okra pods is classified as a very strong antioxidant.

This study uses male rats (Rattus novergicus), aged 8-12 weeks weighing 150-200 grams, with a total of 31 rats. Therapy was conducted by putting rats in a chamber that emits an electric field every day for 1 hour for 28 days. To search for the optimum frequency of the treatment, frequency variations of 15, 30, 60, and 120 kHz were given. The electrical charge distribution of the air in the room was then measured, whereas mice blood sugar levels were also measured to reveal the effect of the treatment on cell tolerance to sugar, and to come up with the measurement of insulin levels at the end of the treatment. This study was done in vivo using male mice (strain BALB/C, 3 to 4 months old, 30 to 40g). The normal control group (KN) and the diabetic groups (induced by streptozotocin). The diabetic group was divided into 9 groups: three diabetic control groups (KD), three alginate treatment groups (KAI), and three treatment groups of alginate-okra (Abelmoschus esculentus) pods extract (KAIO). The treatment dose was 50 mg/kg body weight. Measurement of fasting blood sugar levels was done before and after streptozotocin injection. The treatment was carried out in three different time periods: 3 days, 7 days, and 14 days. Then, mice skin was cut and proceed for histopathological analysis. Interestingly, we indicated that the administration of okra (Abelmoschus esculentus) pods extract combined with alginate from Sargassum ilicifolium could increase the rate of re-epithelialization of the wound area, increase the number of neutrophils, macrophages, fibroblasts, fibrocytes, as well as increase synthesized collagen. The combined extract was able to improve the healing process of open wounds in diabetic mice. Therefore, it can be concluded that the administration of alginate from Sargassum ilicifolium and okra (Abelmoschus esculentus) pods extract was able to improve open wounds healing in diabetic mice due to the role of antioxidant from polysaccharides and quercetin which acted as anti-hyperglycemic to anti-inflammatory properties.

Okra polysaccharides were obtained by water extraction and ethanol precipitation. Okra polysaccharides with doses of 25, 50, 75, and 100 mg/kg BW were orally administrated to mice with or without Staphylococcus aureus infection. This study was using 35 male Balb/c strain mice. The samples were divided into seven groups, namely KN (normal control), KD (diabetic control), KA (acarbose control), EK (crude extract), NP (non-polar), SP (semi-polar), and EP (polar). VOPE and acarbose suspension were administered orally for 14 days in the STZ-induced diabetic mice. Intraperitoneal injection of multiple low-dose STZ (30mg/kg body weight) induced diabetes daily for five consecutive days. On the 15th day, the mice were terminated. Data of SOD level and tissue glucose tolerance were analyzed by ANOVA, at a = 0.05. This study concluded that the administration of VOPE could increase SOD level and tissue glucose tolerance in the STZ-induced diabetic mice. In conclusion, VOPE is a promising antioxidant agent due to its antihyperglycemic properties.

In this study, we used male mice (Mus musculus) of the BALB/C strain which were divided into 2 groups: the control group (without treatment of mangosteen pericarp extract) and the treatment group. The control group was divided into three: normal control (KN), diabetic control (KD), diabetic control-Metformin HCl (KM). The treatment group (with mangosteen pericarp extract) was divided into 3 groups (P1, P2, and P3) with the dose of 50 mg/kg body weight, 100 mg/kg body weight, and 200 mg/kg body weight, respectively. The induction of diabetes was done with the injection of multiple low-doses of STZ (30 mg/kg of body weight) for 5 consecutive days. Before and after STZ injection, random blood glucose and total cholesterol were measured at 1 st , 7 th , and 14 th day of mangosteen pericarp extract treatments. Treatments were given for 14 days. At 15 th day, SGOT, SGPT and plasma creatinine levels were measured using Pentra C200, while liver and kidney were collected and then processed into histological slides. Interestingly, we found that mangosteen pericarp extract administration was able to reduce random blood glucose, total cholesterol, SGOT, SGPT, plasma creatinine levels, also ameliorates damaged liver hepatocytes and renal proximal tubular cells in diabetic mice significantly. In conclusion, mangosteen pericarp extract is a promising antidiabetic agent due to its anti-hyperglycemic and antioxidant properties.

In this study, we used male BALB/C mice. Mice were divided into two groups: normal control (KN) and streptozotocin-induced diabetic. Streptozotocin (STZ) induction was performed using multiple low doses of 30 mg/kg body weight injected for five consecutive days. The diabetic mice were separated into three subgroups: diabetic control (KD), diabetic mice treated with acarbose (KA), and diabetic mice treated with gamma-mangostin at either 0,5 mg/kg body weight (P1), 1 mg/kg body weight (P2), or 2 mg/kg body weight (P3). Before and after STZ injection, fasting blood glucose and cholesterol level would be observed. Fasting blood glucose and cholesterol level were also measured at the 1 st , 7 th , and 14 th day of gamma-mangostin treatment. Treatment was given for 14 days. On the 15 th day, SGOT and SGPT were measured using a Pentra C 200 Reader, while the liver was collected and processed onto the histological slides. Interestingly, we found that gamma-mangostin was able to reduce the fasting blood glucose, cholesterol, SGOT, SGPT, and ameliorate the damaged hepatocytes in significant diabetic mice. Therefore, we concluded that gamma-mangostin is a promising anti-diabetic agent due to its anti-hyperglycemic and antioxidant activities.

VOPE and acarbose were administered orally for 14 days in streptozotocin-induced type-2 diabetic mice. Mice with type-2 diabetes mellitus were induced by the intraperitoneal injection of multiple low-dose streptozotocin (30 mg/kg body weight) daily for five consecutive days. On 15th day, mice were terminated. The data of fasting blood glucose, serum insulin, and GLUT4 density were analyzed and interestingly it showed that the administration of VOPE could ameliorate the fasting blood glucose, serum insulin, and GLUT4 density. Therefore, it can be suggested that VOPE can be regarded as a promising antidiabetic agent in the streptozotocin-induced diabetic mice.

The levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), malondialdehyde (MDA), superoxide dismutase (SOD), and catalase (CAT) were measured using enzyme-linked immunosorbent assays, and the histology of the liver was evaluated using light microscopy. It can activate granulocyte, monocyte, and macrophage. Administration of crude polysaccharide of C. versicolor against bacterial infection is hypothesized to increase immune response. This study assessed the crude polysaccharides activity of C. versicolor in improving immune response after induction of non-tuberculosis mycobacteria, Mycobacterium fortuitum. Animal model was female mice strain BALB/c aged 8-10 weeks. A crude polysaccharide of C. versicolor was administered before and/or after the bacteria infection for 10 days at a dose of 50 mg/kg body weight. Mice exposure to M. fortuitum was performed twice at a dose of 0.5 Mc. Farland. After crude polysaccharides treatment, both serum and peritoneal fluid were isolated. All data collected were analyzed statistically using ANOVA and Duncan test. Oral administration of crude polysaccharide was found to increase phagocyte number (P<0.05, from crude polysaccharides administration before-after infection), improve phagocytic activity (P<0.05, from crude polysaccharides administration before infection), raise both IFN- and antibody level (P<0.05, from crude polysaccharides administration after and before-after infection), and it caused TNF-α levels to tend to a normal concentration (P>0.05, TNF-α levels after crude polysaccharides administration were relatively the same as controls). Administration of crude polysaccharides of C. versicolor could enhance non-specific immune response, specific immune response, and pro-inflammatory cytokine in mice infected by M. fortuitum. These results suggested that crude polysaccharides of C. versicolor may be applied as an effective immunostimulatory agent.
