Vegetables cultivated in soil irrigated with untreated groundwater and municipal-waste-dominated ... more Vegetables cultivated in soil irrigated with untreated groundwater and municipal-waste-dominated (MWD) stream can elevate the concentration of heavy metals (Cd, Fe, Zn, Hg, Cr, and Ni) in edible parts of the crop, affecting food safety and public health worldwide. This study assessed the quality, sources, and distribution of heavy metals in surface soils, MWD stream and groundwater, and edible tissues of leafy and non-leafy vegetables from a major urban farm in the Sekondi-Takoradi metropolis, Ghana. Human health risk due to exposure to the metals in frequently consumed vegetables were investigated. Indigenous leafy vegetables (Corchorus olitorious and Amaranthus spinosus), exotic leafy vegetables (Lactuca sativa, Brassica oleracea, and Brassica rapa), and non-leafy vegetables (Capsicum annum, Raphanus sativus, Daucus carota, and Allium cepa) were collected from the urban farm. The mean concentration of Cd, Hg, and Fe ranged from 0.008-0.027, 0.001-0.013, and 4.517-36.178 mg/ kg fw in edible parts of non-leafy vegetables, respectively and 0.011-0.035, 0.002-0.011, and 3.617-13.695 mg/ kg fw in exotic or indigenous leafy vegetables. The vegetables were less impacted with the metals if compared to similar vegetables produced from other urban farms, locally and in some countries in Africa, Asia, and Europe. Water resource on the farm were not suitable for vegetable crop irrigation since mean concentration of E. coli (200 cfu/mL), Hg (0.009 mg/L), and Cd (0.019 mg/L) in the MWD stream and 80 % of the groundwater sources exceeded the safe limits recommended by the Food and Agriculture Organization. Geo-accumulation index for each metal in soil was 0, however, enrichment factor indicated a high anthropic enriched soil for Cr and Ni. Principal component analysis-multiple linear regression of the metals in soil identified mixed household waste/ fertilizer, fertilizer, and crustal material as main sources for the heavy metal load in soil for which geogenic sources accounted for 74.3 %. Preferentially, Cd and Hg accumulated in Amaranthus spinosus, Daucus carota, and Corchorus olitorious. The estimated daily intake of each metal in the vegetables were below local and international daily dietary intake levels. At the 95 th percentile concentration of each metal, target hazard quotient and the hazard index was <1 for adult male or female who consume the vegetables. Finally, appropriate agri-horticultural practices must be enforced to mitigate Cd, Ni, Cr, and Hg accumulation in the soil-vegetable system since the metals have profound adverse effect on human health.

The existence of pesticides, such as organochlorine pesticides, parathion-ethyl, methamidophos wh... more The existence of pesticides, such as organochlorine pesticides, parathion-ethyl, methamidophos which is banned globally and some current-use non-banned pesticides of organophosphorus and synthetic pyrethroids in freshwater sources is an ecological and public health concern in many countries, including Ghana. Prompted by this concern, the exposure levels and risk assessment of these pesticides to humans and non-target organisms via groundwater and surface water sources in an agricultural catchment dominated by cocoa crops in the Ankobra Basin, Ghana, were investigated. The individual concentrations of the banned pesticides in the surface water and groundwater samples varied from < LOD to 0.110 μg/L and < LOD to 0.055 μg/L, respectively, while the concentrations of the non-banned pesticides ranged from < LOD to 0.925 μg/L and < LOD to 2 μg/L, respectively. High selectivity and sensitivity is reported in the measurements of xanthine in urine by fast sca... more High selectivity and sensitivity is reported in the measurements of xanthine in urine by fast scan cyclic voltammetry (FSV) with a nanostructured carbon fiber sensor of 3.5 +/- 0.4 mum radius. Fabrication of the sensors for the measurements is described. Fabrication of the nanostructure at the carbon fiber sensor surface exposes surface pores. SEM images confirm the formation of the nanostructure. The results indicate that the nanostructure improves the sensitivity and limit of detection (LOD) in the measurements of xanthine and uric acid. The sensors allow rapid direct measurements of xanthine in 2000-fold diluted xanthinuric urine and of uric acid in 2000-fold diluted normal urine. The sensitivity and the LOD of xanthine is 0.40 +/- 0.02 nA microM(-1) (0.995) and 1 microM, respectively, and 0.99 +/- 0.01 nA microM(-1) (0.998) and 500 nM for uric acid. The concentration of xanthine in 2000-fold diluted xanthinuric urine is 1.6 +/- 0.2 muM from FSV and from HPLC. The concentration of xanthine and uric acid in urine can be determined by pre- or post-calibration of the sensor in buffer or by the method of standard addition.

Background: The ineffectiveness of artesunate and amodiaquine tablets in malaria treatment remain... more Background: The ineffectiveness of artesunate and amodiaquine tablets in malaria treatment remains a health burden to WHO and governments of malaria-endemic countries, including Ghana. The proliferation of illegitimate anti-malarial drugs and its use by patients is of primary concern to international and local drug regulatory agencies because such drugs are known to contribute to the development of the malaria-resistant parasites in humans. No data exist on quality of these drugs in the fishing village communities in Ghana although the villagers are likely users of such drugs. A pilot study on the quality of anti-malarial tablets in circulation during the major fishing season at a malarious fishing village located along the coast of Tema in southern Ghana was determined. Methods: Blisterpacks of anti-malarial tablets were randomly sampled. The International Pharmacopoeia and Global Pharma Health Fund Minilab protocols were used to assess the quality of anti-malarial tablets per blisterpacks allegedly manufactured by Guilin Pharmaceutical Co Ltd, China (GPCL) and Letap Pharmaceuticals Ltd, Ghana (LPL) and sold in chemical sales outlets at Kpone-on-Sea. Ferric chloride and cobaltous thiocyanate tests confirmed the presence of active ingredients in the tablets. A confirmatory test for the active ingredient was achieved with artesunate (ICRS1409) and amodiaquine (ICRS0209) reference standards. A high performance liquid chromatography analysis confirmed the amount of artesunate found in tablets. Results: Based on the International Pharmacopoeia acceptable range of 96/98 to 102% for genuine artesunate per tablet, 10% [relative standard deviation (RSD): 3.2%] of field-selected artesunate blisterpack per tablets manufactured by GPCL, and 50% (RSD: 5.1%) of a similar package per tablet by LPL, passed the titrimetric test. However, 100% (RSD: 2.2%) of amodiaquine blisterpack per tablet by GPCL were found to be within the International Pharmacopeia acceptable range of 90 to 110% for genuine amodiaquine in tablet, whilst 17% of a similar package per tablet by LPL failed spectrophotometric testing. Conclusion: Inadequate amounts of artesunate and amodiaquine detected in the tablets suggest that both pharmaceutical companies may not be following recommended drug formulation procedures, or the active pharmaceutical ingredients might have been degraded by improper storage conditions. Thus, drugs being sold at Kpone-on-Sea, Ghana may likely be classified as substandard drugs and not suitable for malaria treatment.

Rosmarinic acid (RA) is an important antioxidant polyphenol that is found in a variety of spices ... more Rosmarinic acid (RA) is an important antioxidant polyphenol that is found in a variety of spices and herbs, including Ocimum canum Sims (locally called eme or akokobesa in Ghana). Aqueous extracts from the leaves of O. canum are used as an antidiabetic herbal medicine in Ghana. Analytical thin-layer chromatography was used to examine the composition of the polyphenols in leaf extracts. The polyphenol content in the aqueous and methanol extracts from the leaf, as determined by the Folin-Ciocalteu method, were 314 and 315 mg gallic acid equivalent/g leaf sample, respectively. The total flavonoid concentration as determined by the aluminum(III) chloride method was 135 mg catechin equivalent/g leaf sample. Highperformance liquid chromatography coupled to an electrospray Quadrupole time-of-flight mass spectrometer was also used to determine the polyphenol fingerprint profile in the leaf extracts of O. canum. Although the average RA concentration in the O. canum leaf extracts from Ghana was 1.69 mg/g dry weight (reported values range from 0.01 to 99.62 mg/g dry weight), this polyphenol was still a prominent peak in addition to caffeic acid derivatives. In Ghana anti-malaria herbal medicines or products are used to compliment commercial drugs in tre... more In Ghana anti-malaria herbal medicines or products are used to compliment commercial drugs in treatment and prevention of Plasmodium falciparum infections. In this study, four common aqu eous based anti-malaria herbal products (coded HEB, KFE, MDM and NIB) which are used by Ghanaian population from pharmacy/herbal stores in the Madina area, Accra were blindly and ran domly sampled for cadmium (Cd), arsenic (As) and Lead (Pb) analysis using Atomic Absorption Spectrophotometry technique. Arsenic concentrations were 1.087 lg/mL (108.7%), 1.027 lg/mL (102.7%), 0.330 lg/mL (33.0%) and 0.274 lg/mL (27.4%) in MDM, KFE, NIB and HEB respectively. Arsenic concentration determine d in MDM and KFE were above the maximum permissible limit of 1.0 ppm determine d by WHO/FAO. Cadmium concentration in each of the four products as well as lead concentration in KFE, NIB and HEB were below the detection limit of <0.002 mg/mL (Cd) and <0.005 mg/mL (Pb) respectively. The maximum permissible limits for Pb and Cd determined by WHO/FAO are 10.0 ppm and 0.3 ppm respectively. Thus, random assessment on the safety of some ready-to-u se aqueous based anti-malaria herbal products on the market is necessary to prevent public health hazards associated with consuming these plant extracts. Although lead and cadmium concentration in the anti-malaria herbal products were below the maximum permissible limits, their cumulative effect on the health of an individual which consume recommen ded volume of not less than 1000 mL for effective malaria parasite clearance cannot be ignored.

Total As content and the As species distribution in water and sediments from the Kwabrafo stream,... more Total As content and the As species distribution in water and sediments from the Kwabrafo stream, a major water body draining the Obuasi gold mining community in southwestern Ghana, have been investigated. Total As content was determined by instrumental neutron activation analysis (INAA). Ion-pair reverse phase high-performance liquid chromatography-neutron activation analysis (HPLC-NAA) was used for speciation of As species. Solid phase extraction with phosphate buffer was used to extract soluble As species from lyophilized sediment. The mass balance after phosphate extraction of soluble As species in sediment varied from 89 to 96 %. Compositionally appropriate reference material International Atomic Energy Agency (IAEA)-Lake Sediment (SL)-1 was used to check the validity of INAA method for total As determination. The measured values are in good agreement with the IAEA recommended value and also within the 95 % confidence interval. The accuracy of the measurement in terms of relative deviation from the IAEA recommended value was ±0.83 %. &amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;quot;In-house&amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;quot; prepared As(III) and As(V) standards were used to validate the HPLC-INAA method used for the As species determination. Total As concentration in the water samples ranged from 1.15 to 9.20 mg/L. As(III) species in water varied from 0.13 to 0.7 mg/L, while As(V) species varied from 0.79 to 3.85 mg/L. Total As content in sediment ranged from 2,134 to 3,596 mg/kg dry mass. The levels of As(III) and As(V) species in the sediment ranges from 138 to 506 mg/kg dry mass and 156 to 385 mg/kg dry mass, respectively.

2,8-Dihydroxyadenine (2,8-DHA) was identified by high-performance liquid chromatography with ultr... more 2,8-Dihydroxyadenine (2,8-DHA) was identified by high-performance liquid chromatography with ultraviolet detection as a major metabolite in the supernatant of endothelial cells of the pulmonary artery (PAECs) and aorta (AECs), in addition to hypoxanthine, xanthine, uric acid, and uracil. Under normoxic, hypoxic, and hyperoxic conditions, the concentrations of all the identified metabolites change with time, marking the response of endothelial cells to stress, as a result of changes in cellular metabolism. Thus, the metabolites can serve as stress markers, and their concentrations can indicate the type and the level of cell stress. The results verify that PAECs adapt to survive oxidative stress of hyperoxia. However, AECs can adapt to hypoxia only for a short time and do not survive prolonged hypoxia. The role of the polyamine synthesis pathway in the formation of the unsalvaged adenine, as a possible source of 2,8-DHA, is discussed.

The mass balance after phosphate extraction of soluble As species in sediment varied from 89 to 96 %. Compositionally appropriate reference material International Atomic Energy Agency (IAEA)-Lake Sediment (SL)-1 was used to check the validity of INAA method for total As determination. The measured values are in good agreement with the IAEA recommended value and also within the 95 % confidence interval. The accuracy of the measurement in terms of relative deviation from the IAEA recommended value was ±0.83 %. &amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;quot;In-house&amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;quot; prepared As(III) and As(V) standards were used to validate the HPLC-INAA method used for the As species determination. Total As concentration in the water samples ranged from 1.15 to 9.20 mg/L. As(III) species in water varied from 0.13 to 0.7 mg/L, while As(V) species varied from 0.79 to 3.85 mg/L. Total As content in sediment ranged from 2,134 to 3,596 mg/kg dry mass. The levels of As(III) and As(V) species in the sediment ranges from 138 to 506 mg/kg dry mass and 156 to 385 mg/kg dry mass, respectively.

2,8-Dihydroxyadenine (2,8-DHA) was identified by high-performance liquid chromatography with ultr... more 2,8-Dihydroxyadenine (2,8-DHA) was identified by high-performance liquid chromatography with ultraviolet detection as a major metabolite in the supernatant of endothelial cells of the pulmonary artery (PAECs) and aorta (AECs), in addition to hypoxanthine, xanthine, uric acid, and uracil. Under normoxic, hypoxic, and hyperoxic conditions, the concentrations of all the identified metabolites change with time, marking the response of endothelial cells to stress, as a result of changes in cellular metabolism. Thus, the metabolites can serve as stress markers, and their concentrations can indicate the type and the level of cell stress. The results verify that PAECs adapt to survive oxidative stress of hyperoxia. However, AECs can adapt to hypoxia only for a short time and do not survive prolonged hypoxia. The role of the polyamine synthesis pathway in the formation of the unsalvaged adenine, as a possible source of 2,8-DHA, is discussed.
