Insulin-like growth factor I (IGF-I) and its binding proteins (IGFBPs), have a very important rol... more Insulin-like growth factor I (IGF-I) and its binding proteins (IGFBPs), have a very important role in the regulation of fetal growth. These protein factors are produced virtually by all fetal organs since early development and are powerful stimulators of cellular division and differentiation. The objective of this review is discuss about the physiological role of IGF-I and its IGFBPs during pregnancy, the regulation of the biological actions of IGF-I by its IGFBPs, and the alterations of this factors during the intrauterine growth retardation and preeclampsia.

The infection with SARS-CoV-2 impairs the glucose–insulin axis and this contributes to oxidative ... more The infection with SARS-CoV-2 impairs the glucose–insulin axis and this contributes to oxidative (OS) and nitrosative (NSS) stress. Here, we evaluated changes in glucose metabolism that could promote the loss of redox homeostasis in COVID-19 patients. This was comparative cohort and analytical study that compared COVID-19 patients and healthy subjects. The study population consisted of 61 COVID-19 patients with and without comorbidities and 25 healthy subjects (HS). In all subjects the plasma glucose, insulin, 8-isoprostane, Vitamin D, H2S and 3-nitrotyrosine were determined by ELISA. The nitrites (NO2−), lipid-peroxidation (LPO), total-antioxidant-capacity (TAC), thiols, glutathione (GSH) and selenium (Se) were determined by spectrophotometry. The glucose, insulin and HOMA-IR (p < 0.001), 8-isoprostanes, 3-nitrotyrosine (p < 0.001) and LPO were increased (p = 0.02) while Vitamin D (p = 0.01), H2S, thiols, TAC, GSH and Se (p < 0.001) decreased in COVID-19 patients in compar...

The effects of nonglycated bovine serum albumin (BSA) and advanced glycosylation end products of ... more The effects of nonglycated bovine serum albumin (BSA) and advanced glycosylation end products of BSA (AGE-BSA) on vascular responses of control and metabolic syndrome (MS) rats characterized by hypertriglyceridemia, hypertension, hyperinsulinemia, and insulin resistance were studied. Albumin and in vitro prepared AGE-BSA have vascular effects; however, recent studies indicate that some effects of in vitro prepared AGEs are due to the conditions in which they were generated. We produced AGEs by incubating glucose with BSA for 60 days under sterile conditions in darkness and at 37 • C. To develop MS rats, male Wistar animals were given 30% sucrose in drinking water since weanling. Six month old animals were used. Blood pressure, insulin, triglycerides, and serum albumin were increased in MS rats. Contraction of aortic rings elicited with norepinephrine was stronger. There were no effects of nonglycated BSA or AGE-BSA on contractions in control or MS rats; however, both groups responded to L-NAME, an inhibitor of nitric oxide synthesis. Arterial relaxation induced using acetylcholine was smaller in MS rats. Nonglycated BSA and AGE-BSA significantly diminished relaxation in a 35% in the control group but the decrease was similar when using nonglycated BSA and AGE-BSA. This decrease was not present in the MS rats and was not due to increased RAGEs or altered biochemical characteristics of BSA. In conclusion, both BSA and AGE-BSA inhibit vascular relaxation in control artic rings. In MS rats the effect is lost possibly due to alterations in endothelial cells that are a consequence of the illness.

Resveratrol (RSV) and quercetin (QRC) modify energy metabolism and reduce cardiovascular risk fac... more Resveratrol (RSV) and quercetin (QRC) modify energy metabolism and reduce cardiovascular risk factors included in the metabolic syndrome (MetS). These natural compounds upregulate and activate sirtuins (SIRTs), a family of NAD-dependent histone deacetylases. We analyzed the effect of two doses of a commercial combination of RSV and QRC on serum fatty acid composition and their regulation of SIRTs 1–3 and PPAR-γexpression in white adipose tissue. MetS was induced in Wistar rats by adding 30% sucrose to drinking water for five months. Rats were divided into control and two groups receiving the two different doses of RSV and QRC in drinking water daily for 4 weeks following the 5 months of sucrose treatment. Thirty two female Wistar rats were divided into 4 groups: Control (C), MS, MS ovariectomized (Ovx), and MS Ovx plus estradiol (E2). MS was induced by administering 30% sucrose to drinking water for 24 weeks. After sacrifice, intra-abdominal fat was dissected; adipocytes were isolated and lipid peroxidation, non-enzymatic antioxidant capacity, and the activities of Cu-Zn and Mn superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) were determined. There were no significant differences in the activities of Cu-Zn, Mn SOD, CAT, and GPx between the C and MS groups, but in the MS Ovx group there was a statistically significant decrease in the activities of these enzymes when compared to MS and MS Ovx+E2. Changes in aortic vasoreactivity during the postnatal pancreatic critical window, where insulin a... more Changes in aortic vasoreactivity during the postnatal pancreatic critical window, where insulin and glucose, which modify vasoreactivity, are elevated, were studied and compared to those in control and metabolic syndrome (MS) rats. Twelve 21- and 28-day-old rats were used. To develop MS rats, male Wistar animals were given 30% sucrose in drinking water since weaning and used when 6 months old. Glucose and insulin levels were higher during suckling and decreased after weaning, and insulin and triglycerides levels increased in MS rats. Contraction elicited by norepinephrine (NE) was stronger than KCl contraction at all ages. KCl-induced contraction increased with, age being stronger in control rats; it further increased in MS rats. Norepinephrine-induced contraction increased from day 12 to day 28 but stabilized from day 21 to day 28; it was stronger in controls and increased in MS rats. Vasorelaxation to acetylcholine in NE precontracted rings did not change during the neonatal period, being similar to MS rats and lower than in controls. Insulin-induced increase in contraction elicited by KCl increased from day 12 to day 28 and increased from control to MS rats. There is a postnatal critical window in vasoreactivity that might predispose to cardiovascular diseases in adults.

Adicionalmente se exploró, in vitro, la capacidad de secreción de insulina estimulada por glucosa de los islotes de Langerhans de estas ratas hiperandrogenizadas e hiperestrogenizadas. Se encontró que la hiperandrogenización se acompañaba de un deterioro de la sensibilidad a la insulina, con hiperinsulinemia, que no se corresponde con un aumento de la capacidad de secreción de insulina de los islotes de Langerhans. The present work provides information about magnesium (Mg) physiology, Mg supplementation during ... more The present work provides information about magnesium (Mg) physiology, Mg supplementation during normal pregnancy, and the use of Magnesium Sulfate MgSO4 in preeclampsia and eclampsia. Intestinal absorption, renal excretion and cellular distribution of Mg are described. In addition, this review includes several functions in which Mg is involved such as bone structure, enzymatical activities, cellular signaling, ionic channel regulation and gene expression. Furthermore, Mg supplementation for the prevention of preterm delivery, low birth weight, and preeclampsia are discussed. Results about the use of MgSO4 as treatment of preeclampsia and eclampsia are also revised. Finally, the effects of Mg upon the synthesis, secretion and/or action of vasoactive factors involved in blood pressure and uteroplacental flux regulation are described.

The quantity and site of accumulation of adipose tissue is of great importance for the physiopathology of this disease. the aim of this study was to assess the effect of a high carbohydrate diet on adipose tissue distribution. male Wistar rats, control (CONT) and high sucrose diet (HSD; 30% sucrose in their drinking water), were monitored during 24 weeks and total energy and macronutrient intake were estimated by measuring daily average consumption. A bioelectrical impedance procedure was performed at 22 weeks of treatment to assess body compartments and systolic arterial blood pressure was measured. Serum was obtained and retroperitoneal adipose tissue was collected and weighed. HSD ingested less pellets and beverage, consuming less lipids and proteins than CONT, but the same amount of carbohydrates. Retroperitoneal adipose tissue was more abundant in HSD. This phenomenon is accelerated in conditions where glucose concentration is chronically high, as it happens in diabetes mellitus. AGE formation is associated with structure-function alterations of proteins such as collagen, and particularly in tissues where these products are accumulated. Se estudió el efecto de una sobredosis de enantato de testosterona y de benzoato de estradiol en ... more Se estudió el efecto de una sobredosis de enantato de testosterona y de benzoato de estradiol en ratas machos prepúberes, sobre el comportamiento de la glucemia y la insulinemia in vivo, durante una prueba de tolerancia a la glucosa. Adicionalmente se exploró, in vitro, la capacidad de secreción de insulina estimulada por glucosa de los islotes de Langerhans de estas ratas hiperandrogenizadas e hiperestrogenizadas. Se encontró que la hiperandrogenización se acompañaba de un deterioro de la sensibilidad a la insulina, con hiperinsulinemia, que no se corresponde con un aumento de la capacidad de secreción de insulina de los islotes de Langerhans. La evidencia sugiere que muchos de estos efectos son mediados por la glucosilacion no enzimatica (o glicacion) de proteinas que lleva a la formacion de los llamados productos finales de glucosilacion avanzada (AGEs por sus siglas en ingles). Este proceso se acelera en condiciones en que la concentracion de glucosa se mantiene elevada por tiempos prolongados, como ocurre en la diabetes mellitus, alterando la estructura y funcion de proteinas como la colagena, asi como de los tejidos en los cuales estos productos se acumulan. Se considero que la castracion se acompanaba de un incremento en la sensibilidad a la insulina, sin deterioro de la capacidad de secrecion de insulina de los islotes. The present work provides information about magnesium (Mg) physiology, Mg supplementation during ... more The present work provides information about magnesium (Mg) physiology, Mg supplementation during normal pregnancy, and the use of Magnesium Sulfate MgSO4 in preeclampsia and eclampsia. Intestinal absorption, renal excretion and cellular distribution of Mg are described. In addition, this review includes several functions in which Mg is involved such as bone structure, enzymatical activities, cellular signaling, ionic channel regulation and gene expression. Furthermore, Mg supplementation for the prevention of preterm delivery, low birth weight, and preeclampsia are discussed. Results about the use of MgSO4 as treatment of preeclampsia and eclampsia are also revised. Finally, the effects of Mg upon the synthesis, secretion and/or action of vasoactive factors involved in blood pressure and uteroplacental flux regulation are described.
