Chemical markers for the quality control of herbal medicines: an overview
Selection of chemical markers is crucial for the quality control of herbal medicines, including authentication of genuine species, harvesting the best quality raw materials, evaluation of postharvesting handling, assessment of intermediates and finished products, and detection of harmful or toxic ingredients. Ideal chemical markers should be the therapeutic components of herbal medicines. However, for most herbal medicines, the therapeutic components have not been fully elucidated or easily monitored. Bioactive, characteristic, main, synergistic, correlative, toxic and general components may be selected. This article reviews the effective use of chemical markers in the quality control of herbal medicines including the selection criteria considering the roles and physicochemical factors which may affect the effective use of chemical markers.

Many countries are registering their standardized herbal drugs with proven clinical efficacy, safety and as dietary supplements. But India is unable to exploit the World market due to unsatisfactory system of quality control which necessitates the establishment of standards for herbal single drugs and compound formulations. In order to ensure international community to use AYUSH drugs with confidence and without compunction, developing acceptable quality control standards of herbal medicines aims to ensure their consistency, safety and efficacy with good manufacturing practices is very much essential. Study of chemical markers is applicable to many research areas, including authentication of genuine species, search for new resources or substitutes of raw materials, optimization of extraction and purification methods, structure elucidation and purity determination. Systematic investigations using chemical markers may lead to discoveries and development of new drugs.

Different chromatographic and electrophoretic techniques commonly used in the instrumental inspection of herbal medicines (HM) are first comprehensively reviewed. Chemical fingerprints obtained by chromatographic and electrophoretic techniques, especially by hyphenated chromatographies, are strongly recommended for the purpose of quality control of herbal medicines, since they might represent appropriately the "chemical integrities" of the herbal medicines and therefore be used for authentication and identification of the herbal products. Based on the conception of phytoequivalence, the chromatographic fingerprints of herbal medicines could be utilized for addressing the problem of quality control of herbal medicines. Several novel chemometric methods for evaluating the fingerprints of herbal products, such as the method based on information theory, similarity estimation, chemical pattern recognition, spectral correlative chromatogram (SCC), multivariate resolution, etc. are discussed in detail with examples, which showed that the combination of chromatographic fingerprints of herbal medicines and the chemometric evaluation might be a powerful tool for quality control of herbal products.

The efficacy and safety of any pharmaceutical product is determined by the compounds (desired and undesired) which it contains. The purpose of quality control is to ensure that each dosage unit of the drug product delivers the same amount of active ingredients and is, as far as possible, free of impurities. As herbal medicinal products are complex mixtures which originate from biological sources, great efforts are necessary to guarantee a constant and adequate quality. By carefully selecting the plant material and a standardized manufacturing process the pattern and concentration of constituents of herbal medicinal products should be kept as constant as possible as this is a prerequisite for reproducible therapeutic results.

In order to achieve the comprehensive quality control of Chinese herbal medicine (CHM), the conventional practice of selecting a single marker for testing has been gradually replaced by the determination of multiple active components based on the characteristics of the synergistic interaction of CHM and applicability of sophisticated analytical techniques. So the different chemical, phytochemical tests, analytical techniques are used for determining the quality aspects of the herbal materials in the herbal pharmaceuticals. The present review is focused on the various quality control tests for herbal drugs such as organoleptic evaluation, chemical evaluation, foreign matter, stability determination, analytical evaluation, etc. we have also discussed the benefits of quality control of herbal medicines. Phytochemical standardization of herbal materials involves establishing consistent levels of one or more active ingredients or markers. It ensures the authenticity and quality of herbal materials, extracts, and medicinal products. We successfully applied the Herbal chemical Markers ranking system (Herb MaRS) originally proposed for quality assurance of complex herbal products, to establish markers for controlling the quality of herbal raw materials. The use of products of natural origin has remarkably increased in the global market, because of consumer becoming more health-conscious herbal medicines are considered safe by consumers and these product are generally less expensive than allopathic medicines.
