Identifikasi Serovar Penyebab Leptospirosis pada Anjing di Yogyakarta

Leptospirosis is a zoonotic disease of global concern, and is caused by pathogenic serovar Leptospira interrogans. Canine Leptospirososis is widespread worldwide, dogs can act as incidental hosts or maintenance hosts for various serovars. The purpose of this research was to identify leptospire serovars that infect healthy and suspected leptospirosis dogs in Yogyakarta. A total of 56 dogs (36 healthy dogs and 20 suspect leptospirosis dogs) sera were taken from cephalica vein as much as 3 ml. Sera were examined for leptospirosis with Microscopic Aglutination Test (MAT) which conducted at the Research Center for Veterinary Science, Bogor. Microscopic Aglutination Test carried out on various Leptospire serovar, namely: Ichterohaemorrhagiae, Javanica, Celledoni, Ballum, Pyogenes, Cynopeteri, Rachmati, Australis, Pomona, Canicola, Grippotyphosa, Bataviae, Hardjo, and Tarrasovi. Rat is a rodent (rodensia) which cannot be separated from parasitic organism attacks the ectoparasites (fleas). In the presence of fleas plague focus areas need to watch out, for no increase in cases of plague (outbreak). Pest is a zoonosis in rat that can be transmitted to humans through the bite of fleas Xenopsylla cheopsis containing Yersinia pestis. Boyolali District is one of the plague focus areas in Central Java. Three important sources of transmission of leptospirosis are the environment contaminated with leptospira, the presence of rats and presence of livestock as reservoirs. This study aims to determine the reservoir that plays a role in the leptospirosis transmission in increased case areas of leptospirosis in the Pagedangan Ilir Village, Kronjo Sub-district, Tangerang. Rat traping was done twice. Each trapping was done two times, in consecutive days. Livestock sampling was also conducted in the study area. Rat and livestock serum specimens were collected for leptospirosis detection using MAT, while rat kidneys were collected for PCR test. Leptospirosis detection was performed by PCR and MAT. The results of the first and second rat traping showed relative density of 17% and 20% in the houses, as well as 14% and 10% in outdoor area. Rat traped consist of two species, Rattus norvegicus and Rattus tanezumi. The prevalence of pathogenic leptospira in rat (with PCR assay) was 33% in the first trap and 26% in the second trap. The MAT test showed a positive 14% on R. norvegicus in the first trap, but negative in the second trap. While livestock did not show PCR positive, but one serological positive sample (by MAT) with low titers (1:20) was found against the serovar Robinsoni.
Rodents are animals that have a high adaptability and they can multiply rapidly in all kinds of places. The existence of rats to humans may be adversely affected as they become agricultural pests and spread infectious diseases such as plague and leptospirosis. Various methods of rat controls have been carried out both traditionally and modernly. This study aims to explore the behavior of people in an attempt to control and its factors that influence the ways to do it. The research was conducted using qualitative design with descriptive analysis. The data collection was done by in-depth interviews and focus group discussion with research location in Nanggulan District, Kulon Progo Regency. Leptospirosis adalah penyakit zoonosis, yang disebabkan oleh Leptospira interrogans. Kejadian leptospirosis pada anjing bervariasi menurut wilayah dan musim, dan dianggap sebagai penyakit menular yang muncul pada manusia. Gejala klinis leptospirosis pada anjing sangat bervariasi, beberapa anjing tanpa menunjukkan gejala, dengan gejala ringan, dan yang lain melanjut menjadi penyakit yang parah sampai kematian. Penelitian ini bertujuan untuk mengetahui kasus leptospirosis pada anjing di Propinsi Daerah Istimewa Yogyakarta, dan mengidentifikasi serovar yang menginfeksinya. Sebanyak 20 ekor anjing tanpa gejala, dengan gejala ringan, sampai gejala parah leptospirosis diambil serumnya guna pemeriksaan Microscopic Agglutination Test (MAT) yang dilaksanakan di Balai Besar Penelitian Veteriner (BBLitvet), Bogor. Sejarah vaksinasi dicatat sebagai bahan pertimbangan dalam menginterpretasi hasil pemeriksaan MAT. Hasil penelitian menunjukkan bahwa tiga dari 20 sampel positif leptospirosis. Satu sampel positif terhadap serovar Bataviae, satu sampel positif terhadap serovar Bataviae dan Tarrasovi, dan satu sampel lagi positif terhadap serovar Bataviae, Tarrasovi, Ichterohaemorrhagiae, Canicola, Celledoni, Pyrogenes, Cynopteri, dan Rachmati. Anjing yang positif leptospirosis menunjukkan gejala klinis yang sama berupa anoreksia, kelemahan, dan demam. Dari hasil penelitian ini dapat disimpulkan bahwa terdapat kasus leptospirosis pada anjing di Yogyakarta yang penyebabnya dominan oleh Leptospira interrogans serovar Bataviae.

Bantul is one of leptospirosis endemic areas which has been reported the cases every year and the presence of rats is an important risk factor relating to leptospirosis incidence in Bantul. Therefore, the role of rats as reservoir was examined in three types of ecosystems: forest, non-forest, and coastal ecosystems. Rat trapping was carried out using 100 single livetraps which were distributed in 9 locations: 2 points in the forest, 5 points in the non-forest, and 2 points in the coastal ecosystem. The rats were identified and their kidneys were collected and preserved in 70% alcohol medium. Leptospira were detected in the kidney by using PCR method. A number of 196 rats were obtained during the study. Most of them were commensal rats obtained from settlements ecosystems. Determinasi Serovar Bakteri Leptospira Pada Reservoir DI Kabupaten Banyumas

Leptospirosis is an infectious disease caused by pathogenic Leptospira. Leptospirosis transmitted to human through direct contact with body fluids of infected animals or indirectly through contaminated puddles . The prevalence of leptospirosis in Banyumas tends to increase for 3 years. The purpose of this study was to determine the leptospira serovar in reservoir to prove of a current infection. Surveys was conducted using single live traps for three consecutive days, determination of leptospira serovar was conducted using Microscopic Aglutination Test (MAT). Data analysis was performed by univariate and presented in tables and graphs. The results showed that the trapped animals consisted of Rattus tanezumi (70.6%) and Suncus murinus (29.4%) with 6.5% succsess trap. Rattus tanezumi were dominantly caught inside the house (51%) than outside the house (49%). Female rats were dominantly caught (66.7%) than male rats (33.3%). Suncus murinus and Rattus tanezumi shown a titer of 1/100 to be infected with L.icterohaemorrhagiae , L.javanica and L.cynopteri which are pathogenic Leptospira in humans. Efforts are needed to improve community participation in preventing tranmission of leptospirosis by avoiding contact with contaminated water and soil. For people who are risk of exposure to infected animal should wear protective clothes or footwear.

Kasus Leptospirosis di Kota Semarang selalu mengalami peningkatan. Puskesmas Bandarharjo adalah Puskesmas yang wilayahnya merupakan zona rawan Leptospirosis. Tahun 2015, di wilayah Puskesmas Bandarharjo ditemukan 22 kasus Leptospirosis, meningkat dari tahun sebelumnya yaitu 14 kasus pada tahun 2014. Kasus Leptospirosis tersebut paling banyak terjadi di Kelurahan Tanjung Mas. Fakta ini menempatkan Puskesmas Bandarharjo tertinggi kasus Leptospirosis di kota Semarang. Wilayah ini termasuk endemis, karena wilayah ini merupakan daerah rawan banjir dan merupakan daerah yang sering terdampak rob (kawasan rob). Banyak genangan air pada musim penghujan maupun akibat rob menyebabkan banyak selokan-selokan yang menggenang, sampah-sampah menumpuk yang menjadi tempat berkembangbiaknya tikus.
