This study was made to predict the weights and percentages of retail cuts in Holstein bull carcas... more This study was made to predict the weights and percentages of retail cuts in Holstein bull carcasses. The study material were 47 Holstein bull (mean slaughter age: 20.57±0.606 months) carcasses. Hot carcass weight (HCW, kg), ribeye area (REA, cm 2) and fat thickness (FT, cm) were used to predict weights and percentages of retail cuts. Bull carcasses were divided into 12 retail cuts and grouped as total retail cuts (TRC), first degree retail cuts (FRC) and second degree retail cuts (SRC). Regression analysis was made (stepwise method) to predict weights and percentages of TRC, FRC and SRC. Slaughter weight (SW), dressing percentage (DP), HCW, REA and FT were determined as average 544.550±6.776 kg; 55.75±0.169%; 303.600±3.948 kg; 85.538±1.978 cm 2 and 0.310±0.023 cm, respectively. All predicted models (F) were found significantly (P<0.001; P<0.01) in the analysis. As a result, it was found that HCW is most important predictor in predicting the weights and percentages of retail cuts. HCW and REA explained for 94.5% (R 2) of the variation of dependent variable (YTRC) in the predicted model for weights of total retail cuts (TRCkg). REA and HCW explained for 21.7% (R 2) of the variation of dependent variable (YTRC) in the predicted model for percentages of total retail cuts (TRC%).

This study aimed to evaluate the antimicrobial activity of methanol, acetone, and aqueous extracts of endemic Sideritis galatica plant collected from Afyonkarahisar. For this purpose, the effect of different concentrations (2.5%, 5%, 10%, 20%) of these extracts on L. monocytogenes, B. cereus, S. aureus, E. faecalis, E. coli, E. coli O157, S. typhimurium, C. albicans were investigated by the disk diffusion method. Methanol extract showed antimicrobial activity against all of the tested microorganisms. Also, acetone and aqueous extracts showed antimicrobial activity against all of the tested microorganisms except E. faecalis (for acetone extract), E. coli and E. coli O157 (for aqueous extract). Consequently, S. galatica as an endemic plant showed antimicrobial activity especially at high concentrations against tested microorganisms. Endemik Sideritis galatica bitkisinin antimikrobiyal etkisinin araştırılması ÖZET: Bitkisel çaylar Türkiye'de yaygın olarak tüketilmektedir. Bu çalışmada Afyonkarahisar'dan toplanan endemik Sideritis galatica bitkisinin metanol, aseton ve sulu ekstraktlarının antimikrobiyal aktivitesi değerlendirildi. Bu amaçla, bu ekstraktların farklı konsantrasyonlarının (% 2,5, % 5, % 10, % 20) L. monocytogenes, B. cereus, S. aureus, E. faecalis, E. coli, E. coli O157, S. typhimurium, C. albicans disk difüzyon yöntemi ile araştırıldı. Metanol ekstraktı test edilen tüm mikroorganizmalara karşı antimikrobiyal aktivite gösterdi. Ayrıca, aseton ve sulu ekstraktlar, E. faecalis (aseton ekstraktı için), E. coli ve E. coli O157 (sulu ekstrakt için) hariç test edilen tüm mikroorganizmalara karşı antimikrobiyal aktivite gösterdi. Sonuç olarak, endemik bir bitki olarak Sideritis galatica, test edilen mikroorganizmalara karşı özellikle yüksek konsantrasyonlarda antimikrobiyal aktivite gösterdi.

This study has aimed to detect Brucella abortus and Brucella melitensis in fresh cheese (Coban Cheese, 100-mold), traditional Afyon Tulum Cheese (Aged Skin Bag Cheese) (50-mold) and sheep-lamb tulum (50-mold) samples provided from Afyonkarahisar province. Conventional Culture Method has been used in the isolation and identification of B. abortus and B. melitensis. In the case study, Brucella spp. has been detected at the rates of 9% (2% B. abortus, 7% B. melitensis) in the analyzed fresh cheese samples and 6% (2% B. abortus, 4% B. Melitensis) in Afyon Tulum Cheese. However, Brucella spp. has not been detected in analyzed sheep-lamb tulum samples. Consequently, it has been accepted that fresh cheese and Afyon Tulum Cheese samples are contaminated with B. abortus and B. melitensis which impose a high risk for the public health.

In this study, survival of Listeria monocytogenes 1/2b was investigated in Ayran samples. For that purpose, Ayran samples produced from yoghurt (Group A and B) or directly form milk (Group C and D) were contaminated with 1 % concentration of 7 (A1, B1, C1 and D1 samples) or 5 (A2, B2, C2 and D2 samples) log cfu/mL of L. monocytogenes. So, eight different samples of Ayran were produced and stored at 4 oC (Group A and C) or 20 oC (Group B and D) for 21 days. According to the obtained results, there was no significant change in the number of viable cells of L. monocytogenes 1/2b in samples A1, A2, C1 and C2 after 21 days of the storage period. Bu bakteriler mastitisli hayvanlar, hijyenik olmayan sagim ekipmanlari, transport veya depolama esnasinda cig sute bulasabilmektedir. Yapilan bu calismada Afyonkarahisar ilinden toplanan 100 manda ve 100 inek sutu olmak uzere toplam 200 cig sut numunesi materyal olarak kullanildi. Toplanan sut numuneleri ISO 11290-1 yontemi kullanilarak Listeria spp. varligi yonunden analiz edildi. Analiz edilen 100 cig manda sut orneginin 1’inde (%1) ve 100 cig inek sut orneginin 4’unde (%4) Listeria spp. klasik kultur metodu ile izole edildikten sonra Microbact 12L test kiti kullanilarak identifiye edildi. Ayrica izole ve identifiye edilen Listeria turleri PCR ile onaylandi. This study has aimed to evaluate the prevalence of the brucellosis in sheep and cattle in a Rural Area of Western Anatolia, (Turkey). Methods: In this scope, blood samples were obtained from 1065 sheep and 756 cattle while milk samples were obtained from 500 sheep and 500 cows in Afyonkarahisar provincial area. Findings: As a result, it has been detected that brucellosis positivity in sheep was13.9% with Rose Bengal (RB), 6.39% with Standard Tube Agglutination (STA); while the prevalence in cattle was 4.37% with RB, 3.56% with STA. Taze salatalar ve salata yapiminda kullanilan sebzeler, cig tuketildigi icin patojen bakteriler i... more Taze salatalar ve salata yapiminda kullanilan sebzeler, cig tuketildigi icin patojen bakteriler ile kontamine olmasi durumunda insan sagligi acisindan ciddi riskler olusturabilmektedir. Bu amacla yapilan calismada, Afyonkarahisar ilinde tuketime sunulan taze marul orneklerinin mikrobiyolojik kalitesi incelendi. Calisma kapsaminda, market ve pazarlardan toplanan 70 adet taze marul ornegi Esherichia coli O157 ve Listeria monocytogenes varligi yonunden analize alindi. E. coli O157 ve L. monocytogenes analizleri sirasiyla ISO 16654 ve ISO 11290-1’e gore gerceklestirilmistir. Analiz sonucunda, 70 adet taze marul orneginin 2 (%2,86)’sinde E. coli O157 ve bir (%1,43) tane taze marul orneginde ise L. monocytogenes , tespit edildi. Sonuc olarak, incelenen marul orneklerinin patojen bakterileri icerdigi ve mikrobiyolojik kalitesinin dusuk seviyede oldugu belirlenmistir. Isil islem olmadan tuketilen sebzelerde E. coli O157 ve L. monocytogenes gibi patojen bakterilerin bulunmasi saglik acisindan ciddi bir risk olusturmaktadir. Bu nedenle, mikrobiyolojik kaliteyi artirmak icin etkili kontrol onlemlerinin uygulanmasi gerekmektedir.

Consumption of such foods contributes to the protection of our health and strengthening of the immune system. This research was conducted to determine the consumption status of probiotic products and the level of knowledge consumers have about probiotic products. It was found in this survey that 51.7% of the participants were females, and 48.1% were males, and ages ranged between 18 and 65. It was also found that while 31.4% of males were informed about probiotic products, 61.1% of females were informed. As regards the educational statuses, it was seen that level of knowledge about probiotic products significantly increased with increasing educational level from primary school level to higher levels. Background: Screening of transmission routes and routine control of the food for foodborne-pathog... more Background: Screening of transmission routes and routine control of the food for foodborne-pathogens are vital in terms of public health. In this study, we aimed to investigate and evaluate the presence of E. coli O157:H7 strains and toxins in the cattle meat samples collected from different markets and butchers. Methods: We collected 116 raw minced cattle beef samples from the supermarkets and the butcher stores. We used bacterial culture-based conventional isolation methods as recommended by the CDC and FDA determination of STEC in the minced cattle beef samples. Then we used PCR to detect stx genes in sorbitol negative E. coli. This way, we indirectly demonstrated the presence of the stx genes in meat samples. Additionally, we used an agglutination test for the detection of E. coli O157:H7. Results: E. coli O157-suspected isolates were found in 17 (14.6%) out of 116 raw minced meat samples examined with tests. STEC stx toxin gene was found in 14 (12.06%) of the sorbitol-negative E. coli isolates tested with real-time PCR method. There was no statistical difference between samples collected from markets and butchers according to STEC stx toxin gene positivity rate. Latex agglutination method performed very poor results in suspected strains compared the PCR results (p < 0.05). Conclusions: Meat products sold in markets and butchers carry low but similar risks for infections and epidemics in our region. In the studies that evaluate the presence of the STEC, agglutination methods cannot be trusted alone and, therefore, this test should be combined with at least one of the conventional microbiological or molecular methods.

Bisphenol A (BPA) is one of the most produced chemicals in the world and has been widely employed... more Bisphenol A (BPA) is one of the most produced chemicals in the world and has been widely employed in the food industry. Continuous and widespread exposure to BPA through drinking water and food leads to health concerns for humans. This study evaluated the effects of boron (B) on BPA-mediated oxidative stress in male Wistar albino rats. Rats were equally divided into 5 groups; corn oil was given orally to the control group; 25 mg kg −1 of BPA dissolved in corn oil was given orally to the second group. All other groups received the same dose of BPA and different doses of B (5, 10, and 20 mg kg −1 per day, respectively) orally for 30 days. The administration of BPA significantly decreased glutathione levels and increased malondialdehyde levels in rat tissues. Furthermore, BPA treatment reduced the catalase and superoxide dismutase activities in tissues and erythrocytes. Also, mRNA expression levels of TNF-α, IL-1β, and IL-6 in the brain, liver, and testes of rats were augmented, whereas IL-10 was decreased with BPA treatment. Besides, BPA treatment adversely altered biochemical parameters and caused damage to the cell integrity of rat tissues. However, B administration reversed BPA-induced alterations in rat tissues in a dose-dependent manner. In this study, survival of Listeria monocytogenes 1/2b was investigated in Ayran samples. For that purpose, Ayran samples produced from yoghurt (Group A and B) or directly form milk (Group C and D) were contaminated with 1 % concentration of 7 (A1, B1, C1 and D1 samples) or 5 (A2, B2, C2 and D2 samples) log cfu/mL of L. monocytogenes. So, eight different samples of Ayran were produced and stored at 4 oC (Group A and C) or 20 oC (Group B and D) for 21 days. According to the obtained results, there was no significant change in the number of viable cells of L. monocytogenes 1/2b in samples A1, A2, C1 and C2 after 21 days of the storage period. Bu amacla yapilan calismada, Afyonkarahisar ilinde tuketime sunulan taze marul orneklerinin mikrobiyolojik kalitesi incelendi. Calisma kapsaminda, market ve pazarlardan toplanan 70 adet taze marul ornegi Esherichia coli O157 ve Listeria monocytogenes varligi yonunden analize alindi. E. coli O157 ve L. monocytogenes analizleri sirasiyla ISO 16654 ve ISO 11290-1’e gore gerceklestirilmistir. Analiz sonucunda, 70 adet taze marul orneginin 2 (%2,86)’sinde E. coli O157 ve bir (%1,43) tane taze marul orneginde ise L. monocytogenes , tespit edildi. Sonuc olarak, incelenen marul orneklerinin patojen bakterileri icerdigi ve mikrobiyolojik kalitesinin dusuk seviyede oldugu belirlenmistir. Isil islem olmadan tuketilen sebzelerde E. coli O157 ve L. monocytogenes gibi patojen bakterilerin bulunmasi saglik acisindan ciddi bir risk olusturmaktadir. Bu nedenle, mikrobiyolojik kaliteyi artirmak icin etkili kontrol onlemlerinin uygulanmasi gerekmektedir.

Consumption of such foods contributes to the protection of our health and strengthening of the immune system. This research was conducted to determine the consumption status of probiotic products and the level of knowledge consumers have about probiotic products. It was found in this survey that 51.7% of the participants were females, and 48.1% were males, and ages ranged between 18 and 65. It was also found that while 31.4% of males were informed about probiotic products, 61.1% of females were informed. As regards the educational statuses, it was seen that level of knowledge about probiotic products significantly increased with increasing educational level from primary school level to higher levels. Background: Screening of transmission routes and routine control of the food for foodborne-pathog... more Background: Screening of transmission routes and routine control of the food for foodborne-pathogens are vital in terms of public health. In this study, we aimed to investigate and evaluate the presence of E. coli O157:H7 strains and toxins in the cattle meat samples collected from different markets and butchers. Methods: We collected 116 raw minced cattle beef samples from the supermarkets and the butcher stores. We used bacterial culture-based conventional isolation methods as recommended by the CDC and FDA determination of STEC in the minced cattle beef samples. Then we used PCR to detect stx genes in sorbitol negative E. coli. This way, we indirectly demonstrated the presence of the stx genes in meat samples. Additionally, we used an agglutination test for the detection of E. coli O157:H7. Results: E. coli O157-suspected isolates were found in 17 (14.6%) out of 116 raw minced meat samples examined with tests. STEC stx toxin gene was found in 14 (12.06%) of the sorbitol-negative E. coli isolates tested with real-time PCR method. There was no statistical difference between samples collected from markets and butchers according to STEC stx toxin gene positivity rate. Latex agglutination method performed very poor results in suspected strains compared the PCR results (p < 0.05). Conclusions: Meat products sold in markets and butchers carry low but similar risks for infections and epidemics in our region. In the studies that evaluate the presence of the STEC, agglutination methods cannot be trusted alone and, therefore, this test should be combined with at least one of the conventional microbiological or molecular methods.

Continuous and widespread exposure to BPA through drinking water and food leads to health concerns for humans. This study evaluated the effects of boron (B) on BPA-mediated oxidative stress in male Wistar albino rats. Rats were equally divided into 5 groups; corn oil was given orally to the control group; 25 mg kg −1 of BPA dissolved in corn oil was given orally to the second group. All other groups received the same dose of BPA and different doses of B (5, 10, and 20 mg kg −1 per day, respectively) orally for 30 days. The administration of BPA significantly decreased glutathione levels and increased malondialdehyde levels in rat tissues. Furthermore, BPA treatment reduced the catalase and superoxide dismutase activities in tissues and erythrocytes. Also, mRNA expression levels of TNF-α, IL-1β, and IL-6 in the brain, liver, and testes of rats were augmented, whereas IL-10 was decreased with BPA treatment. Besides, BPA treatment adversely altered biochemical parameters and caused damage to the cell integrity of rat tissues. However, B administration reversed BPA-induced alterations in rat tissues in a dose-dependent manner.
