Als Schüler seines Vaters (1754–1827) war er 1809 als Assistent Robert Smirkes am Wiederaufbau des Covent-Garden-Theaters beschäftigt und studierte von 1810 bis 1817 die antike Architektur in Italien, Griechenland und Kleinasien. Er beteiligte sich an der Erforschung des Aphaiatempels in Ägina und fand mit Carl Haller von Hallerstein und Jakob Linckh die später nach München verkauften Figuren der Giebelfelder. Auch nahm er an den Ausgrabungen bei Phigalia teil und ließ den Fries des Apollontempels bei Bassae nach London schaffen. Dieser ist im Britischen Museum zu sehen. 1817 eröffnete er, nach London zurückgekehrt, sein eigenes Atelier, das ziemlich erfolgreich war. In der Folge lieferte er unter anderem die Entwürfe für die Church of Holy Trinity in Bristoler Stadtteil Hotwells (1829), für die Gebäude der Bank von England in Plymouth (1835), Bristol (1844–1847), Manchester (1845) und Liverpool (1845–1848), das Ashmolean Museum in Oxford (1839–1845) und die Erweiterung des Fitzwilliam Museums in Cambridge (1848). 1841 wurde Cockerell als auswärtiges Mitglied in die Académie des Beaux-Arts aufgenommen. On returning to London, he set up a successful architectural practice. Appointed Professor of Architecture at the Royal Academy of Arts, he served in that position between 1839 and 1859. He wrote many articles and books on both archaeology and architecture. In 1848, he became the first recipient of the Royal Gold Medal.
