The main challenge to probiotics, during their passage through the gastrointestinal tract, are the acidic gastric secretions of the stomach, and the bile salts released into the duodenum. The survival of the strains, in this phase, is strongly influenced by the food used for their delivery. This work is part of a project studying the development of novel food processes, based on the use of chestnuts from cultivar "Castagna di Montella". In detail, the effect of indigestible chestnut fiber and of chestnut extract on the viability of selected lactic acid bacteria strains was evaluated. Among 28 cultures, twelve strains were selected, on the basis of tolerance to low pH values and bile salts, and submitted to exposition to simulated gastric or bile juice in presence of chestnut extract with or without immobilization in chestnut fiber. The presence of chestnut extract proved to play a significant role on the gastric tolerance improvement of lactobacilli. The recorded protective effect could not be simply related to the starch or reducing sugars content. RP-HPLC demonstrated that in the chestnut flour, there are one or more hydrophobic peptides or oligopeptides, which specifically offer a marked resistance to simulated gastric juice, albeit present at low concentration. These beneficial effects proved to be dependent by the cultivar used to produce the flour.

Emesis encompasses the forceful expulsion of the contents of stomach via the mouth or sometimes the nose.
The adverse effects of currently available anti-emetic agents potentiate the natural product researchers to explore the
natural anti-emetics with fewer side effects. The presented communication constitutes a review on anti-emetic effect of
two hundred and forty five plants belonging to seventy-eight families found in different parts of the world. It also
outlined the anti-emetic effect of plant extracts and isolated secondary metabolites studied through a variety of animal
models of emesis. The reported anti-emetic plants in different countries and cultures and the scientific studies on extracts
may help in the identification of promising single chemical compound(s) that may be used as a potential leads for
developing safe anti-emetic agents in future. Moreover the reported secondary metabolites having the same effect may
open the door for the search of same secondary metabolites from other natural sources. This review will provide useful
information for the discovery of natural anti-emetic compounds and fill the gaps in knowledge.

The early Burdigalian (MN3) plant assemblage of the Güvem area (northwestern Central Anatolia) is preserved in lacustrine sediments of the Dereköy pyroclastics. Its age is well constrained by radiometric dates of basaltic rocks bracketing the pyroclastics, making the Güvem flora one of the extremely few precisely dated early Miocene floras in the Mediterranean region. The rich assemblage of impression fossils comprises ferns and fern allies (2 species), gymnosperms (12 spp.) and angiosperms (129 spp.). Ilex miodipyrena sp. nov. is described as a new fossil-species. The most diverse families in the assemblage are the Fagaceae with 12 taxa and the Fabaceae with 12 leaf morphotypes and one fruit taxon. Aquatic plants are represented by seven taxa, riparian (including palms) and swamp forest elements by >35 taxa, and lianas by three taxa (Smilax spp., Chaneya). The relatively large number of aquatic and riparian/swamp elements is congruent with the rich fish, amphibian and reptile record of the Güvem area. Another characteristic feature of the plant assemblage is the presence of various lobed leaves which show similarities with modern species of different families (e.g. Alangium, various Malvales). Trees and shrubs growing on well-drained soils and forming closed-canopy and open-canopy forests are the most diversified group (>70 taxa). In terms of number of specimens in the collection and based on field observations, by far the most abundant leaf fossils belong to evergreen oaks of Quercus drymeja and Q. mediter-ranea and to various types of foliage that cannot be assigned to a particular extant or extinct genus of Fagaceae. These sclerophyllous trees must have covered vast areas surrounding the wetlands that developed during the early Miocene in the Güvem Basin. Based on a recent reassessment of the ecology and taxonomic affinity of these trees, they are considered to reflect humid temperate climatic conditions but with a brief drier season during the winter months. These forests are more similar to the laurel forests of the southeastern United States and those stretching in a narrow belt south of the Himalayas to eastern central China. The large number of Fabaceae may indicate the presence of warm subtropical environments but this is difficult to assess, as they are known for having wide ecological ranges today and in the past. All in all, a larger part of the plant taxa point to forested vegetation. This is in agreement with previous palynological studies which detected only small amounts of her-baceous and grass pollen. Open patches of vegetation may have been restricted to river banks and to rocky areas in a volcanic landscape. The biogeographic patterns detected for the early Miocene of the Güvem assemblage are manifold; most taxa are widespread Northern Hemispheric elements. A substantial part of the species migrated from Asia into Europe during the (late) Paleogene and reached Anatolia during the early Miocene (Fagus, Paliu-rus, Chaneya, Ailanthus, Quercus kubinyii, Davallia haidingeri, Acer angustilobum, A. palaeosaccharinum). Fewer taxa may have been in Anatolia before they migrated to Europe (e.g. Nerium, Smilax miohavanensis, Quercus sosnowskyi). Finally, very few taxa are Anatolian endemics (e.g. Ilex miodipyrena).

The uppermost Eocene Florissant Formation, Rocky Mountains, Colorado, has yielded numerous insect, vertebrate and
plant fossils. Three previous comprehensive palynological studies investigated sections of lacustrine deposits of the
Florissant Formation and documented the response of plant communities to volcanic eruptive phases but overall found
little change in plant composition throughout the investigated sections. These studies reported up to 150 pollen and spore
phenotypes. In the present paper, we used a taxonomic approach to the investigation of dispersed pollen and spores of the
Florissant Formation. Sediment samples from the shale units containing macrofossils were investigated using light
microscopy (LM) and scanning electron microscopy (SEM). The general picture of the palynoflora is in agreement
with previous studies. However, the combined LM and SEM investigation provides important complementary information
to previous LM studies. While a fairly large amount of previous pollen determinations could be confirmed, the purported
taxonomic affinities of several pollen phenotypes need to be revised. For example, pollen referred to as Podocarpus or
Podocarpidites sp. belongs to the Pinaceae Cathaya, Malus/Pyrus actually belongs to Dryadoideae, pollen of the form genus
Boehlensipollis referred to as Proteaceae/Sapindaceae/Elaeagnaceae or Cardiospermum belongs to Sapindaceae but not to
Cardiospermum, and pollen of Persicarioipollis sp. B with previously assumed affinities to Polygonaceae actually belongs to
Thymelaeaceae. Pandaniidites and one type of Malvacipollis cannot be linked with Pandanaceae and Malvaceae. A few taxa
are new records for Florissant (Ebenaceae: Diospyros; Menispermaceae; Trochodendraceae: Tetracentron). In general,
SEM investigations complement the LM palynological studies and improve the identification of dispersed pollen and
spores and enable integration of data from dispersed fossil pollen into a wide range of comparative morphological,
taxonomic, evolutionary, biogeographic and phylogenetic studies.

Oaks of Quercus Group Ilex are emblematic components of the Mediterranean landscapes and the full extent of their diversity in a geographic context is still poorly assessed. In order to detail differentiation patterns within Group Ilex and to illuminate causes and circumstances that underlie the distribution of its lineages, we examined plastome differentiation in the four species currently representing this oak group across the Mediterranean Basin (Quercus ilex, Quercus coccifera, Quercus aucheri, Quercus alnifolia). Plastid DNA variation was explored with three markers on 124 individuals and spatial analyses were performed with phylogenetic and landscape genetics approaches. A strong geographic sorting, entirely decoupled from species boundaries, was found. Spatial coincidences with phylogenetic splits highlights the presence of four geographic regions (the Middle East, the Aegean region, the central and the western Mediterranean) characterised by different degrees of isolation. Distinct sub-regional structures were identified. Inter-and intra-lineage divergence patterns appear to reflect the complex orogeny of the Mediterranean region and Pleistocene climatic oscillations, while a few widespread haplotypes might represent the remnants of the ancestral Group Ilex range. Regional differentiation in this group is recognised to be triggered by a number of factors, including disruption of the original range, incomplete lineage sorting, repeated phases of asymmetrical introgression and isolation. The key role played by some Mediterranean regions on the range establishment and dynamics of this oak group is discussed.

The purpose of the study was the identification of the mechanisms of resistance to Dryocosmus kuriphilus Yasumatsu in the hybrid-resistant cultivar 'Bouche de Bétizac' (Castanea sativa  Castanea crenata). Larvae and eggs of the insect are found in the buds of this cultivar at the end of winter, but there is no gall development after budburst. The hypothesis of the presence of a hypersensitive reaction (HR) in the buds was tested using diaminobenzidine (DAB) to detect H 2 O 2 and by Real Time PCR (RT-PCR) to evaluate the expression of a germin-like protein gene. HR in plants is elicited by the production of reactive oxygen compounds, such as H 2 O 2 , and results in the programmed cell death. The DAB test was applied to buds of 'Bouche de Bétizac' and of the susceptible cultivar 'Madonna' (C. sativa) at different stages of budburst. The DAB staining produced brown areas in the swelling buds of 'Bouche de Bétizac', indicating the presence of H 2 O 2. On the contrary, all uninfested buds, as well as the infested buds of 'Madonna', appeared whitish. Papers report that germin and germin-like proteins (GLP) with oxalate oxidase activity are discrete markers of stress-responsive gene products. A strong expression of the chestnut GLP gene was detected by RT-PCR at bud swelling in infested 'Bouche de Bétizac' buds but not in 'Madonna' ones. The results support the hypothesis of the occurrence of an HR in 'Bouche de Bétizac' as response to the cynipid infestation, resulting in cell and larvae death.

Background Chronological information on the composition and structure of agrocenoses and detailed features of land cover referring to specific areas are uncommon in ethnobotanical studies, especially for periods before the XIX century. The aim of this study was to analyse the type of crop or the characteristics of soil cover from the XVI century to the present. Methods This diachronic analysis was accomplished through archival research on the inventories of the Parish of St. Mary and those of the Municipality of Pignone and from recent surveys conducted in an area of eastern Liguria (Italy). Results Archival data revealed that in study area the primary means of subsistence during the last five centuries, until the first half of the XX century, was chestnuts. In the XVIII and XIX centuries, crop diversification strongly increased in comparison with previous and subsequent periods. In more recent times, the abandonment of agricultural practices has favoured the re-colonisation of mixed woodland or cluster-pine woodland. Conclusion Ancient documents in the ecclesiastic or municipal inventories can be a very useful tool for enhancing the knowledge of agricultural practice, as well as of subsistence methods favoured by local populations during a particular time and for reconstructing land use change over time.

Climate change has affected the rates of chilling and heat accumulation, which are vital for flowering and production, in temperate fruit trees, but few studies have been conducted in the cold-winter climates of East Asia. To evaluate tree responses to variation in chill and heat accumulation rates, partial least squares regression was used to correlate first flowering dates of chestnut (Castanea mollissima Blume) and jujube (Zizyphus jujube Mill.) in Beijing, China, with daily chill and heat accumulation between 1963 and 2008. The Dynamic Model and the Growing Degree Hour Model were used to convert daily records of minimum and maximum temperature into horticulturally meaningful metrics. Dryocosmus kuriphilus is one of the most damaging pests of Castanea spp. Behavioral, chemical, and electrophysiological investigations were employed to examine the role of plant volatiles for host location by this thelytokuos cynipid. Y-tube olfactometer bioassays showed that adult wasps are significantly attracted by C. sativa twigs with at least 1-hr-old mechanical damage. Odors of undamaged host seedlings, intact twigs, and twigs with a fresh mechanical damage were not attractive. Wasps were repelled by plant materials of the non-host Prunus laurocerasus. Fourteen compounds, mainly general green leaf volatiles, were identified in the head-space of attractive host plant twigs by gas chromatography coupled to mass spectrometry. All compounds elicited dose-dependent antennal responses in adult wasps. A synthetic blend comprising all identified compounds in the same ratio as in the attractive host source induced significant positive responses in Y-tube olfactometer bioassays. The study gives a basis for future identification of host plant attractants that could contribute to semiochemical-based monitoring and management practices of this pest.

An ongoing investigation of the middle Miocene (Sarmatian) palynoflora from the Lavanttal Basin continues to show that it contains an extremely rich assemblage of angiosperm taxa. The Fagales to Rosales pollen record documented here contains 34 different taxa belonging to the Betulaceae (Alnus, Two of the pollen types represent extinct genera, Trigonobalanopsis and Cedrelospermum, and are also reported for the first time from the Lavanttal Basin along with pollen of Rhamnaceae and Prunus. The different types of Quercus pollen are now affiliated with Groups Cerris, Cyclobalanopsis, Ilex and Quercus/Lobatae based on sculpturing elements observed using scanning electron microscopy (SEM). Köppen signatures of potential modern analogues of the fossil Fagales and Rosales suggest a subtropical (Cfa, Cwa) climate at lower elevation and subsequent subtropical to temperate climate with altitudinal succession (Cfa → Cfb/Dfa→ Dfb; Cwa → Cwb → Dwb) in the Lavanttal area during accumulation of the palynoflora. Most of the fossil taxa have potential modern analogues that can be grouped as nemoral and/or merido-nemoral vegetation elements, and the diversity of Fagales indicates a varying landscape with a high variety of niches.

Modern lineages of the beech family, Fagaceae, one of the most important north-temperate families of woody flowering plants, have been traced back to the early Eocene. In contrast, molecular differentiation patterns indicate that the Fagus lineage, Fagoideae, with a single modern genus, evolved much earlier than the remaining lineages within Fagaceae (Trigonobalanoideae, Castaneoideae, Quercoideae). The minimum age for this primary split in the Fagaceae has been estimated as 80 ± 20 Ma (i.e. Late Cretaceous) in recently published, time-calibrated phylogenetic trees including all Fagales. Here, we report fagaceous fossils from the Campanian of Wyoming (82–81 Ma; Eagle Formation [Fm]), the Danian of western Greenland (64–62 Ma; Agatdal Fm), and the middle Eocene of British Columbia (ca 48 Ma; Princeton Chert), and compare them to the Fagaceae diversity of the recently studied middle Eocene Hareøen Fm of western Greenland (42–40 Ma). The studied assemblages confirm that the Fagus lineage (= Fagoideae) and the remainder of modern Fagaceae were diverged by the middle Late Cretaceous, together with the extinct Fagaceae lineage(s) of Eotrigonobalanus and the newly recognised genus Paraquercus, a unique pollen morph with similarities to both Eotrigonobalanus and Quercus. The new records push back the origin of (modern) Fagus by 10 Ma and that of the earliest Fagoideae by 30 Ma. The earliest Fagoideae pollen from the Campanian of North America differs from its single modern genus Fagus by its markedly thicker pollen wall, a feature also seen in fossil and extant Castaneoideae. This suggests that a thick type 1 foot layer is also the plesiomorphic feature in Fagoideae although not seen in any of its living representatives. The Danian Fagus pollen of Greenland differs in size from those of modern species but is highly similar to that of the western North American early Eocene F. langevinii, the oldest known beech so far. Together with the Quercus pollen record, absent in the Campanian and Danian formations but represented by several types by the middle Eocene, this confirms recent dating estimates focussing on the genera Fagus and Quercus, while rejecting estimates from all-Fagales-dated trees as too young. The basic Castaneoideae pollen type, still found in species of all five extant genera of this putatively paraphyletic subfamily, represents the ancestral pollen type of most (modern) Fagaceae (Trigonobalanoideae, Castaneoideae, Quercoideae).

Infusions and decoction of chestnut tree flowers have been used for different medical purposes, but their phytochemical profile and antioxidant activity are still mostly unknown. Herein, decoctions and infusions of flowers from the two most appreciated chestnut cultivars (longal and judia) in Trás-os-Montes, Portugal, were prepared and characterized with regard to their content in free sugars, organic acids, and phenolic compounds, such as flavonoids and hydrolyzable tannins, and their antioxidant activity. Overall, the decoction of the cultivar judia was the sample with both the highest quantity of flavonoids and antioxidant activity. The phenolic compound with the highest abundance in all samples was trigalloyl-HHDP-glucoside, followed by pentagalloyl glucoside. The sample with the highest quantity of total phenolic compounds was judia infusion, closely followed by longal decoction, which also gave the highest quantities of ellagitannins. Regarding sugars and organic acids, the profiles were more similar. These results corroborate ancestral claims of the health benefits of infusions and decoctions of chestnut flowers.

As the third part of an ongoing investigation of middle Miocene palynofloras in the Yatağan Basin (YB), southwestern Anatolia, the palynofloras of the Salihpaşalar lignite mine in the main YB were studied. Seven types of algal spores, aplanospores/zygospores or cysts, six types of lycophyte and fern spores, 12 types of gymnosperm pollen and 90 types of angiosperm pollen were identified. Of a total of ca. 140 plant taxa described from the YB, over 10% are confined to the Salihpaşalar assemblage. Differences between coeval palynofloras of the Sekköy Member might reflect changing or prograding depositional environments. A number of rare accessorial taxa reflect these local differences: Pilularia, Valeriana, Drosera and Persicaria aff. amphibia only occur at Salihpaşalar and are typical of shallow water or temporary ponds associated with a lake shore. Apart from this, all the palynofloras, originating from the lignite seams and overlying limnic limestones (uppermost Turgut and Sekköy Member), of the YB are strongly indicative of extensive woody vegetation with a dominance of diverse Fagaceae and Pinaceae. In addition, a list comparing the well-documented YB palynomorphs to morphologically similar palynomorphs of published late early to middle Miocene plant assemblages of western Anatolian was compiled. Such a comparison reveals that in many instances different taxon names have been used to denote the same taxa. Hence, resolving these synonymies is a prerequisite of any meaningful comparison of palynofloras in the region.

Head-space solid-phase microextraction (HS-SPME) coupled to gas-chromatography-mass spectrometry was developed and applied to obtain the volatile aromatic fingerprints of three typical Italian wines, Valpolicella, Amarone and Recioto, all produced in the restricted geographical area of Valpolicella (Veneto, Italy) with the same grape cultivars within the regulations of a rigid disciplinary of production. Differences between the three typologies are mainly linked to the different withering times to which grapes are subjected before vinification, which strongly influences the concentration and the development of volatile aroma compounds. A total of 22 different wines (7 Valpolicella, 10 Amarone and 5 Recioto) were characterised in terms of aromatic volatile profile with the aim to distinguish the different products and to evaluate the possibility to differentiate the same product from different brands. For the chemometric evaluation of the data one-way analysis of variance (ANOVA), principal component analysis (PCA) and hierarchical cluster analysis (HCA) were tested. All the chemometric tools employed allow to differentiate between the three products. More intriguing is the ability of the chemometric approach to differentiate between the same product (Amarone, Recioto) from different winery, thus showing the potential of this approach to characterize the brand-dependent typicality of wines, which is usually related to subtle technological differences which nevertheless have strong influences on the organoleptic characteristics of the products.

Consumers' demand for innovative probiotic products has recently increased. In previous studies, chestnuts were evaluated as substrate for the growth of lactobacilli and chestnut extract was found to enhance acid tolerance of probiotic strains. The main objectives of this study were to evaluate the suitability of chestnut extract as carrier for spray drying of two probiotic Lactobacillus rhamnosus strains and to develop a probiotic food chestnut based. The optimal settings for the spray-drying processes were defined and the loads of undamaged cells in the dried powders were quantified. Spray-dried cultures were incorporated into an anhydrous basis for chestnut mousse developed ad hoc. In this form, viable cells remained stable over 10(8) CFU g(-1) during a 3 months long storage at 15°C. Sensorial analysis did not highlighted significant differences (P < 0·05) in preference between probiotic-supplemented and control mousses. Results suggest that chestnut mousse, a food product naturally rich in antioxidant compounds, may represent an excellent carrier for probiotics delivering. To authors' knowledge, this is the first information on the survival of lactobacilli in an anhydrous basis for dessert.

The fossilized birth–death (FBD) model can make use of information contained in multiple fossils representing the same clade, and we here apply this model to infer divergence times in beeches (genus Fagus), using 53 fossils and nuclear sequences for all nine species. We also apply FBD dating to the fern clade Osmundaceae, with about 12 living species and 36 fossils. Fagus nuclear sequences cannot be aligned with those of other Fagaceae, and we therefore use Bayes factors to choose among alternative root positions. The crown group of Fagus is dated to 53 (62–43) Ma; divergence of the sole American species to 44 (51–39) Ma and divergence between Central European F. sylvatica and Eastern Mediterranean F. orientalis to 8.7 (20–1.8) Ma, unexpectedly old. The FBD model can accommodate fossils as sampled ancestors or as extinct or unobserved lineages; however, this makes its raw output, which shows all fossils on short or long branches, problematic to interpret. We use hand-drawn depictions and a bipartition network to illustrate the uncertain placements of fossils. Inferred speciation and extinction rates imply approximately 5Â higher evolutionary turnover in Fagus than in Osmundaceae, fitting a hypothesized low turnover in plants adapted to low-nutrient conditions. Tricholoma matsutake produces commercially valuable, yet uncultivable, mushrooms (matsutake) in association with pines in the Far East and Scandinavia and with both pines and oaks in the foothills of Tibet. Other matsutake mushrooms, such as Tricholoma anatolicum from the Mediterranean regions and Tricholoma magnivelare and Tricholoma sp. from the North Pacific Coast area of Canada and North America as well as Mexico, respectively, are associated with pines or oaks in their natural habitats. Tricholoma bakamatsutake and Tricholoma fulvocastaneum from Asia produce moderately valuable matsutake mushrooms and are solely associated with Fagaceae in nature. In this study, we demonstrate for the first time that matsutake mushrooms from Scandinavia, Mediterranean regions, North America, and Tibet form ectomycorrhizae with Pinus densiflora similar to the Far East T. matsutake. In general, worldwide T. matsutake and the symbionts of Pinaceae colonize the rhizospheres of P. densiflora as well as T. matsutake isolated from the host plant. However, T. fulvocastaneum and T. bakamatsutake formed a discontinuous Hartig net and no Hartig net, respectively, and colonized to a lesser extent as compared to T. matsutake. The data suggest that conifer-associated matsutake mushrooms in their native habitat will associate symbiotically with the Asian red pine.

Leaf and cupule/nut remains belonging to Fagus (beech) have previously been reported from three plant-bearing formations on Iceland, the 15 Ma Selárdalur-Botn Formation, the 13.5 Ma Dufansdalur-Ketilseyri Formation, and the ca. 9-8 Ma SkarWstrfnd-Mó kollsdalur Formation, and have traditionally been assigned to various species. In this paper, we study Fagus leaves and cupules from the Miocene of Iceland based on unpublished and newly collected material and assess their taxonomic position. Leaves from Selárdalur (15 Ma) and Tafla (13.5 Ma) are described as a new species, Fagus friedrichii, on grounds of unique combinations of morphological features such as the large elliptical leaves with densely spaced secondary veins, the attenuate leaf apex, and the consistently craspedodromous venation.

Consumers' demand for innovative probiotic products has recently increased. In previous studies, chestnuts were evaluated as substrate for the growth of lactobacilli and chestnut extract was found to enhance acid tolerance of probiotic strains. The main objectives of this study were to evaluate the suitability of chestnut extract as carrier for spray drying of two probiotic Lactobacillus rhamnosus strains and to develop a probiotic food chestnut based. The optimal settings for the spray-drying processes were defined and the loads of undamaged cells in the dried powders were quantified. Spray-dried cultures were incorporated into an anhydrous basis for chestnut mousse developed ad hoc. In this form, viable cells remained stable over 10(8) CFU g(-1) during a 3 months long storage at 15°C. Sensorial analysis did not highlighted significant differences (P < 0·05) in preference between probiotic-supplemented and control mousses. Results suggest that chestnut mousse, a food product naturally rich in antioxidant compounds, may represent an excellent carrier for probiotics delivering. To authors' knowledge, this is the first information on the survival of lactobacilli in an anhydrous basis for dessert.

The genus Castanea (Fagaceae) is widely distributed in the deciduous forests of the Northern Hemisphere. The striking similarity between the Xoras of eastern Asia and those of eastern North America and the diVerence in chestnut blight resistance among species has been of interest to botanists for a century. To infer the biogeographical history of the genus, the phylogeny of Castanea was estimated using DNA sequence data from diVerent regions of the chloroplast genome. Sequencing results support the genus Castanea as a monophyletic group with Castanea crenata as basal. The three Chinese species form a strongly supported sister clade to the North American and European clade. A unique westward expansion of extant Castanea species is hypothesized with Castanea originating in eastern Asia, an initial diversiWcation within Asia during the Eocene followed by intercontinental dispersion and divergence between the Chinese and the European/North American species during the middle Eocene and a split between the European and the North American species in the late Eocene. The diVerentiation within North America and China might have occurred in early or late Miocene. The North America species are supported as a clade with C. pumila var. ozarkensis, the Ozark chinkapin, as the basal lineage, sister to the group comprising C. pumila var. pumila, the Allegheny chinkapin, and Castanea dentata, the American chestnut. Morphological evolution of one nut per bur in the genus may have occurred independently on two continents.

BACKGROUND: Farm animals in intensive farming systems are frequently exposed to oxidative stress, which demands adequate antioxidant protection. The aim of this study was to evaluate the antioxidant potential of different concentrations of Sweet chestnut wood extract (SCW; 0.75, 1.5 and 3 g kg −1) in case of n-3 PUFA-induced oxidative stress in young pigs. RESULTS: The highest concentration (3 g kg −1) of SCW decreased malondialdehyde excretion in urine by 31.7%, but had no effect on plasma malondialdehyde. A linear trend towards decrease of urine isoprostanes iPF 2α-VI was observed with the addition of SCW. All three concentrations of SCW efficiently protected blood lymphocytes from DNA damage and lowered plasma alanine aminotransferase levels. The antioxidative and antigenotoxic effect of 3 g SCW kg −1 feed was comparable to the effect of 90.4 mg kg −1 of added vitamin E. CONCLUSION: The results from this study show that, besides being known as antihelmintic, antimicrobial and antiviral agent, Sweet chestnut wood extract could also be considered as a promising natural antioxidant in animal nutrition.

Monoamine oxidase (MAO) inhibitors are generally used in the treatment of depressive disorders and some neurodegenerative illnesses, such as Parkinson's disease and Alzheimer's disease. The aim of this preliminary study was to investigate the MAO [MAO (E.C.1.4.3.4)] inhibiting effect of various apitherapeutic products, such as chestnut honey, pollen and propolis. Extracts' MAO inhibition was measured using peroxidase-linked spectrophotometric assay in enzyme isolated from rat liver microsomes, and the values are expressed as the inhibition concentration (IC 50 ) causing 50% inhibition of MAO. The antioxidant activity of the bee products was also determined in terms of total phenolic content (TPC) and ferric reducing/ antioxidant power in aquatic extracts. All samples exhibited substantial inhibition of MAO, propolis having the highest. Inhibition was related to samples' TPCs and antioxidant capacities. These results show that bee products possess a sedative effect and may be effective in protecting humans against depression and similar diseases.

It has been suggested that pollen ornamentation can be used to distinguish infrageneric groups in Quercus. Here, we document pollen morphology of nearly all species of Quercus Group Ilex and show that they share distinctive microrugulate pollen ornamentation. Furthermore, pollen ultrastructure of all six infrageneric groups of Quercus was studied comparatively and evaluated using a phylogenetic framework for Fagaceae. Characteristics of the foot layer, such as its thickness and continuity, and the ratio foot layer to tectum are not stable within infrageneric groups of Quercus. The foot layer in derived groups (Groups Cerris, Quercus, Lobatae) is irregular, discontinuous and thin (type 2 foot layer). Members of Groups Cyclobalanopsis and Ilex either have a regular, continuous, and comparatively thick foot layer (type 1), or a type 2 foot layer. Only Group Protobalanus consistently has a type 1 foot layer and is similar to Trigonobalanus and Colombobalanus among trigonobalanoids, and to castanoids. Because (I) all except for one species of Castanea and Nothofagus have a type 1 foot layer and (II) basal groups in Quercus may have a type 1 foot layer, we suggest that this is the ancestral state in Fagaceae, and that the type 2 foot layer of Fagus is derived.

Dating the subsidence history of the North Atlantic Land Bridge (NALB) is crucial for understanding intercontinental disjunctions of northern temperate trees. Traditionally, the NALB has been assumed to have functioned as a corridor for plant migration only during the early Cenozoic, but recent fi ndings of plant fossils and inferences from molecular studies are challenging this view. Here, we report dispersed pollen of Quercus from Late Miocene sediments in Iceland that shows affi nities with extant northern hemispheric white oaks and North American red oaks. Older (15 to 10 Ma) sediments do not contain pollen of Quercus suggesting it arrived after that time. Pollen from the 9 -8 Ma Hr ú tagil locality is indistinguishable from morphotypes common among white and red oaks. In contrast, pollen from the 5.5 Ma Sel á rgil locality has a tectum that is at present confi ned to North American white and red oaks, indicating a second episode of migration to Iceland. These fi ndings suggest that transatlantic migration of temperate plant taxa may not have been limited by vast areas of sea or by cold climates during the Miocene. Furthermore, our results offer a plausible explanation for the remarkably low degree of genetic differentiation between modern disjunct European and North American oaks.

Soil borne Phytophthora cinnamomi and Phytophthora cambivora are considered the most pathogenic species associated with chestnut (Castanea sativa) decline in Europe. Mapping their incidence and distribution from nursery and plantation soils may offer valuable information for limiting spread. As conventional biological baiting and taxonomic confirmation is generally time consuming, labour, logistically and space intensive, we have focused on the development of a specific touchdown nested multiplex Polymerase Chain Reaction (PCR) approach for the simultaneous detection of both species direct from soil. Pre-existing and novel primers, based on Internal Transcribed Spacer (ITS) sequences, have been evaluated for their specificity and use in a multiplex capacity in various combinations. Coupled to this we have modified a mechanical lysis procedure for DNA extraction from up to 10 g of chestnut under storey soils (ranging from 0.5 to 25 mg DNA g À1 fresh soil). Using serial dilutions and/or polyvinylpolypyrrolidone chromatography purification, both species have been successfully detected, in artificially and naturally infected soils. Levels of assay detection are comparable to other Phytophthora species where PCR based diagnostic systems have been reported. A qualitative evaluation of this approach against conventional baiting is presented.

In a previous study, a rapid acidification of soil solution was observed between 1987 and 1997 in a cryptopodzolic soil in southern Switzerland despite a reduction in acidic deposition. The molar ratio of base nutrient cations to aluminum (BC/Al) in the soil solution was used to assess acidification. The monitoring of the soil solution chemistry was continued at the same site between 1998 and 2003 to find out how long the delay in reaction to reduced deposition would last and whether the BC/Al ratios would recover. The reevaluation of all data collected during the 16-year observation period showed no clear improvement in the BC/Al ratios, except below the litter layer where the ratios greatly increased after 1998. Initial signs of recovery were also detected in the mineral horizons, the ratios stabilizing in the second part of the observation period. Sulfate concentrations decreased significantly below the litter mat in response to decreased S deposition. BC concentrations markedly declined below the litter layer and in the mineral horizons, which was attributed to the depletion of the BC exchangeable pool as a result of continued acidic deposition.

On the north regions of Portugal and Spain, the Castanea sativa Mill. culture is extremely important. The biggest productivity and yield break occurs due to the ink disease, the causal agent being the oomycete Phytophthora cinnamomi. This oomycete is also responsible for the decline of many other plant species in Europe and worldwide. P. cinnamomi and Phytophthora cambivora are considered, by the generality of the authors, as the C. sativa ink disease causal agents. Most Phytophthora species secrete large amounts of elicitins, a group of unique highly conserved proteins that are able to induce hypersensitive response (HR) and enhances plant defense responses in a systemic acquired resistance (SAR) manner against infection by different pathogens. Some other proteins involved in mechanisms of infection by P. cinnamomi were identified by our group: endo-1,3beta-glucanase (complete cds); exo-glucanase (partial cds) responsible by adhesion, penetration, and colonization of host tissues; glucanase inhibitor protein (GIP) (complete cds) responsible by the suppression of host defense responses; necrosis-inducing Phytophthora protein 1 (NPP1) (partial cds); and transglutaminase (partial cds) which inducts defense responses and disease-like symptoms. In this mini-review, we present some scientifically advanced solutions that can contribute to the resolution of ink disease.

Flavonoids have been identified as plant signals and developmental regulators for nodulation in legumes and have long been hypothesized to play a critical role during actinorhizal nodulation. Along with dehydration, the development of insects and microorganisms is the major drawback in chestnut conservation. Irradiation has been regaining interest as an alternative technology to increase food products shelf-life. In the present work, the effects of low dose gamma irradiation on sugars, fatty acids and tocopherols composition of chestnuts stored at 4 ºC for different storage periods (0, 30 and 60 days) was evaluated. The irradiations were performed in a 60 Co experimental equipment, for 1 h (0.27  0.04 kGy) and 2 h (0.54  0.04 kGy). Changes in sugars and tocopherols were determined by high performance liquid chromatography (HPLC) coupled to refraction index (RI) and fluorescence detections, respectively, while changes in fatty acids were analysed by gas-chromatography coupled to flame ionization detection (GC-FID). Regarding sugars composition, storage time proved to have higher effect than irradiation treatment. Fructose and glucose increased after storage, with the corresponding decrease of sucrose. Otherwise, tocopherols content was lower in nonirradiated samples, without a significant influence of storage. Saturated (SFA), monounsaturated (MUFA) and polyunsaturated (PUFA) fatty acids levels were not affected neither by storage nor irradiation. Nevertheless, some individual fatty acids concentrations were influenced by one of both factors, such as the increase of palmitic acid in irradiated samples or the decrease of oleic acid after 60 days of storage. Overall, the assayed irradiation doses seem to be a promising alternative treatment to increase chestnuts shelf-life, without affect the profile and composition in important nutrients.

Circa 1900, a farmer from the eastern US planted 11 American chestnut (Castanea dentata) seeds on a newly established farm near West Salem in western Wisconsin. These trees were very successful, producing a large stand of over 6,000 trees. Since this area is well outside the natural range of chestnut, these trees remained free from chestnut blight until 1987. In the West Salem stand, chestnuts are the dominant species of a mixed forest community, reminiscent of the chestnut-oak ecosystems of pre-1900 Appalachia. To identify putative mycorrhizal associates of chestnut in this unique forest, our approach was twofold: (1) an extensive fruiting body survey was conducted for four seasons that yielded approximately 100 putative mycorrhizal species and (2) a belowground molecular approach was used to generate DNA sequences of the internal transcribed spacer region from ectomycorrhizae. Unexpectedly, chestnut did not appear to be the dominant underground ectomycorrhizalforming plant species. This study highlights the need to identify the plant host species when conducting below-ground molecular-based surveys and provides preliminary identification of ectomycorrhizal fungi associated with a disjunct stand of American chestnut.

Three ascomycetous yeast strains, H-6 T, ZX-15 and ZX-20, isolated from the bark of two tree species of the family Fagaceae collected from different regions of China, formed unconjugated and persistent asci containing two to four globose ascospores. 26S rDNA D1/D2 domain and internal transcribed spacer (ITS) region (including 5.8S rDNA) sequence analysis showed that they were closely related to the currently accepted Saccharomyces species with strong support. Comparisons of the rDNA sequences, electrophoretic karyotypes and physiological characters indicated that the three strains represent a novel species in the genus Saccharomyces. Background1471-2229-9-51: American chestnut (Castanea dentata) was devastated by an exotic pathogen in the beginning of the twentieth century. This chestnut blight is caused by Cryphonectria parasitica, a fungus that infects stem tissues and kills the trees by girdling them. Because of the great economic and ecological value of this species, significant efforts have been made over the century to combat this disease, but it wasn't until recently that a focused genomics approach was initiated. Prior to the Genomic Tool Development for the Fagaceae project, genomic resources available in public databases for this species were limited to a few hundred ESTs. To identify genes involved in resistance to C. parasitica, we have sequenced the transcriptome from fungal infected and healthy stem tissues collected from blight-sensitive American chestnut and blight-resistant Chinese chestnut (Castanea mollissima) trees using ultra high throughput pyrosequencing. Results: We produced over a million 454 reads, totaling over 250 million bp, from which we generated 40,039 and 28,890 unigenes in total from C. mollissima and C. dentata respectively. The functions of the unigenes, from GO annotation, cover a diverse set of molecular functions and biological processes, among which we identified a large number of genes associated with resistance to stresses and response to biotic stimuli. In silico expression analyses showed that many of the stress response unigenes were expressed more in canker tissues versus healthy stem tissues in both American and Chinese chestnut. Comparative analysis also identified genes belonging to different pathways of plant defense against biotic stresses that are differentially expressed in either American or Chinese chestnut canker tissues. Conclusion: Our study resulted in the identification of a large set of cDNA unigenes from American chestnut and Chinese chestnut. The ESTs and unigenes from this study constitute an important resource to the scientific community interested in the discovery of genes involved in various biological processes in Chestnut and other species. The identification of many defenserelated genes differentially expressed in canker vs. healthy stem in chestnuts provides many new candidate genes for developing resistance to the chestnut blight and for studying pathways involved in responses of trees to necrotrophic pathogens. We also identified several candidate genes that may underline the difference in resistance to Cryphonectria parasitica between American chestnut and Chinese chestnut.

In Northeast of Portugal, the macrofungal community associated to chestnut tree (Castanea sativa Mill.) is rich and diversified. Among fungal species, the ectomycorrhizal Pisolithus tinctorius and the saprotroph Hypholoma fasciculare are common in this habitat. The aim of the present work was to assess the effect of the interaction between both fungi on growth, nutritional status, and physiology of C. sativa seedlings. In pot experiments, C. sativa seedlings were inoculated with P. tinctorius and H. fasciculare individually or in combination. Inoculation with P. tinctorius stimulated the plant growth and resulted in increased foliar-N, foliar-P, and photosynthetic pigment contents. These effects were suppressed when H. fasciculare was simultaneously applied with P. tinctorius. This result could be related to the inhibition of ectomycorrhizal fungus root colonization as a result of antagonism or to the competition for nutrient sources. If chestnut seedlings have been previously inoculated with P. tinctorius, the subsequent inoculation of H. fasciculare 30 days later did not affect root colonization, and mycorrhization benefits were observed. This work confirms an antagonistic interaction between ectomycorrhizal and saprotrophic fungi with consequences on the ectomycorrhizal host physiology. Although P. tinctorius is effective in promoting growth of host trees by establishing mycorrhizae, in the presence of other fungi, it may not always be able to interact with host roots due to an inability to compete with certain fungi.

Oligosaccharide synthesis is an important cryoprotection strategy used by woody plants during winter dormancy. At the onset of autumn, starch stored in the stem and buds is broken down in response to the shorter days and lower temperatures resulting in the buildup of oligosaccharides. Given that the enzyme DSP4 is necessary for diurnal starch degradation in Arabidopsis leaves, this study was designed to address the role of DSP4 in this seasonal process in Castanea sativa Mill. The expression pattern of the CsDSP4 gene in cells of the chestnut stem was found to parallel starch catabolism. In this organ, DSP4 protein levels started to rise at the start of autumn and elevated levels persisted until the onset of spring. In addition, exposure of chestnut plantlets to 4°C induced the expression of the CsDSP4 gene. In dormant trees or cold-stressed plantlets, the CsDSP4 protein was immunolocalized both in the amyloplast stroma and nucleus of stem cells, whereas in the conditions of vegetative growth, immunofluorescence was only detected in the nucleus. The studies indicate a potential role for DSP4 in starch degradation and cold acclimation following low temperature exposure during activity-dormancy transition.
